摘  要:目的:建立高效液相色谱-蒸发光散射检测器(HPLC-ELSD)测定黄芪甲苷含量的方法。方法:采用蒸发光散射检测器(ELSD)以黄芪甲苷为对照品对参芪颗粒中黄芪甲苷进行HPLC分析,色谱柱Agilent C18(250 mm×4.6 mm,5μm),流动相乙腈-水(32:68),流速0.8mL/min,柱温30℃,ELSD漂移管温度105℃,载气流速2.5 L/min。结果:黄芪甲苷在30.48μg/mL~304.8μg/mL范围内线性关系良好,回收率为97.64%(,RSD=1.11%)。结论:HPLC-ELSD法具有良好的精密度和重现性,可用于参芪颗粒中黄芪甲苷含量测定的质量控制。The research is to establish the method using the high - performance liquid chromatography - evapora- tive light scattering detection ( HPLC - ELSD) to determine the contents of Astragaloside. ELSD was used to measure the contents of Astragaloside in Shenqi granules on the basis of astragaloside standards for control with HPLC analysis. Other conditions were set as follows : Chromatographic column Agilent C18(250 mm×4.6 mm,5μm), mobile phase of acetonitrile - water ( 32 : 68 ), flow rate 0.8 in mL/min, column temperature at 30 ℃, ELSD drift tube temperature at 105 ℃, flow rate of carrier gas in 2.5 L/min. The Results showed that when as- tragaloside is between 30.48 μg/mL and 304.8 tLg/mL, the linear relationship is fine and the recovery rate is 97.64% (RSD = 1.11% ). HPLC -ELSD is precise and reproducible, and can be used to control the quality of the preparation.
