Effects of Trace Element and Organic Matter on Propagation and Growth of Stevia Rebaudiana Bertoni Shoots in Vitro

摘  要:以甜叶菊试管苗为外植体,研究了MS培养基中微量元素对甜叶菊试管苗增殖、生根、生长及移栽等影响;同时也探讨了培养基中添加马铃薯汁、椰子汁、香蕉泥和水解乳蛋白等对试管苗增殖、生根、生长及移栽的影响。结果表明,微量元素对试管苗生长、增殖、生根产生重要作用,其中硼、锌、铜、铁的影响较为明显,尤其是锌元素是培养基中不可或缺的元素,它不仅影响试管苗的生长,而且对试管苗的移栽也产生较大影响,增加MS培养基中的锌元素的浓度,可促进试管苗的增殖、改善试管苗的质量;培养基中添加马铃薯汁、椰子汁、香蕉泥对试管苗的增殖具有一定的促进作用,其中香蕉泥能明显改进试管苗的质量,试管苗茎杆粗壮、生长旺盛,但添加水解乳蛋白却抑制试管苗的增殖和生长;在试管苗生根阶段,添加有机质作用不明显。the shoots of Stevia Rebaudiana Bertoni were used as explants to study the effects organic matter--juice of potato, coconut milk, paste of banana and lactalbumin Hydrolysate of trace elements and in MS basic medium on shoot propagation, rooting, growth and transplant. The results showed that trace element, for example boron (B), zinc(Zn), copper(Cu) and ferric(Fe) played important roles in Stevia Rebaudiana Bertoni shoots propa- gation, rooting or growth. Most of all, Zn was an indispensable element, which influenced not only shoot growth but also transplant, in the MS medium. Zn element increase in the MS medium could promote shoot proliferation and improve the quality of shoot. The medium added by organic matter---juice of potato, coconut milk, paste of banana could promote shoot proliferation and improve the quality of plantlets too, which got shorter, stronger and more vigorous. However, the MS medium added by lactalbumin Hydrolysate restrained the planflet growth and propagation. Adding organic matter in the medium had not obvious impacts on shoot rooting in vitro culture.
