摘 要:目的建立柿叶槲皮素和山柰素含量测定方法。方法对HPLC方法中的色谱条件进行摸索,并采用正交设计法考察供试品溶液的制备条件。结果色谱条件为:Hypersil ODS2 C18柱(4.6 mm×250 mm,5μm),流动相为甲醇-0.4%磷酸溶液(50∶50),流速1.0 ml/min,检测波长360 nm。样品供试液制备条件为:药材破碎度过60目筛,提取溶剂为甲醇-25%盐酸(4∶1),溶剂加入量10 ml,回流时间1 h。结论建立的HPLC含量测定方法准确可靠,可作为壮药材柿叶中槲皮素和山柰素总量的测定方法。Objective To investigate the method for content determination of quercetin and kaempferol in persimmon leaves. Methods To research the chromatographic conditions of high performance liquid chromatography ( HPLC ), then the orthogonal design was applied to study the samples' preparation conditions. Results Chromatographic condition as follows : Hypersil ODS2 C18 (4.6 mm × 250 mm ,5μm) ,the mobile phase consisted of methanol - 0.4% phosphoric acid(50 : 50) with the flow of 1.0 ml/min,and the detection wavelength was 360 nm. The samples' preparation conditions as follows : The fragmentation degree was 60 mesh, extraction solvent was methanol - 25% hydrochloride (4 : 1 ), solvent inventory amounted to l0 ml, reflux time was 1 hour. Conclusion The HPLC method we established is accurate and reliable, which can be used for determination of quercetin and kaempferol in Zhuang medicine of persimmon leaves.
