摘 要:目的建立紫黄凝胶剂中左旋紫草素的含量测定方法。方法采用HPLC法,色谱柱为Acclaim 120 C18(4.6×250mm,5μm),流动相为甲醇-水-磷酸(78∶22∶0.3,v/v),检测波长为516nm,柱温为35℃。结果在8.56-42.80μg·mL-1之间,左旋紫草素进样量和峰面积具有良好的线性关系,线性方程为y=0.4088x+0.033,r=0.9999;测得紫黄凝胶剂中左旋紫草素最低平均含量为0.0615mg·g-1。结论该方法简便可行,重现性好,可作为紫黄凝胶剂的质量控制方法。OBJECtiVE To establish the determination method of L-Shikonin in Zihuang gel. METHODS The column was Acclaim 120 C18(4.6 x250mm, 5μm), mobile phase was methanol-water-phosphoric acid(78:22 :0.3, v/v), the detection wavelength was 516nm, column temperature was 35°C. RESULTS L-shikonin sample volume and the peak area had a good linear relationship in 8.56-42.80μg. mL-1, linear equation was y = 0. 4088x + 0.033, r = 0.9999; the lowest average concentration of L-Shikonin in Zihuang gel was 0.0615mg.g-1 CONCLUSION The method is simple, feasible, and reproducible and can be used as the quality control method for Zihuang gel.
