摘  要:应用ISSR(inter-simple sequence repeat)分子标记技术对21株不同来源的桦褐孔菌(Inonotusobliquus)菌株进行DNA指纹图谱分析,结果表明:从60条引物中筛选出11条ISSR引物对桦褐孔菌基因组DNA进行了ISSR-PCR扩增,共扩增出DNA谱带183条,其中多态性谱带169条,多态位点百分率为92.3%,遗传相似系数变化范围为0.605~0.880。在遗传相似系数为0.763时,可将21个桦褐孔菌菌株划为6大类群,菌株聚类结果与菌株的地理分布有关。ISSR(inter-simple sequence repeat) molecular markers were used to differentiate 21 Inonotus obliquus wild-type strains originating from different locations worldwide.A total of 183 bands were amplified using 11 ISSR primers(selected from 60 test primers),of which 169 bands were polymorphic.ISSR similarity coefficients ranged from 0.605-0.880,and a dendrogram based on UPGMA(unweighted pair group method with arithmetic average) cluster analysis revealed that the 21 strains clustered into six groups at a similarity coefficient of 0.763.Cluster analysis further demonstrated that the genetic distance between the different strains was highly correlated to the geographical origin.
