摘  要:目的探讨柴陈片抗抑郁作用及其作用机理。方法对小鼠进行强迫游泳、悬尾、高架十字迷宫、爬梯、自主活动测定实验;结合慢性不可预见轻度应激抑郁大鼠模型,研究柴陈片的抗抑郁、抗焦虑作用。结果与空白对照组比较,柴陈片能缩短小鼠强迫游泳及悬尾的不动时间(P〈0.01),增高小鼠开臂次数(OE)和开臂时间(OT)的百分比,减少站立次数(P〈0.05,P〈0.01)。与模型组比较,柴陈片组体质量增长明显,大鼠垂直运动次数和水平运动次数均明显增加,糖水偏爱度明显增高,血清中SOD、GSH—Px活力增加,MDA含量下降,差异均有统计学意义(P〈0.05~0.01);各剂量组的柴陈片均不影响小鼠的自主活动。结论柴陈片能改善行为绝望模型小鼠的抑郁行为,能改善焦虑模型小鼠的矛盾心理和焦虑行为,改善慢性应激大鼠的抑郁症状,不具有中枢兴奋性。改善机体脂质过氧化状态可能其作用机制之一。Objective To explore the antidepressant effect and mechanism of Chaichen tablets. Methods By perform- ing the experiments such as forced swimming, tail suspension, elevated plus maze, step climbing, autonomic activi- ties determination in mice, and by applying chronic unpredictable mild stress-induced depression rat model, we studied the antidepressant and anxiolytic effects of Chaichen tablets. Results Compared with the blank control group, Chaichen tablets shortened the mouse forced swimming immobility time and the immobility time in tail suspension test, increased OE % and OT % in mice, and decreased mouse erection frequency. In chronic unpredictable mild stress-induced depression rat models, body weight, vertical movement and level movement times, sucrose prefer- ence, and serum SOD and GSH-Px activities were significantly increased, and serum MDA content decreased in Chaichen tablets group as compared with the model group. However, Chaichen tablets had no effect on mouse sponta- neous activities. Conclusion Chaichen tablets have certain effect on improving depression behavior in mice model of despair, relieving ambivalence and anxiety behavior in anxiety mice model, relieving the depression symptoms in mild stress-induced depression rats, but have no central excitability. Improving the status of lipid peroxidation status may be one of its therapeutic mechanisms.
