摘 要:目的观察2型糖尿病大鼠肌肉组织中过氧化物酶体增殖物激活受体2(PPAR-γ2)的表达,探讨2型糖尿病的病变机制。方法 SD大鼠随机分为正常对照组和模型组,各10只。模型组高脂饲料喂养2个月后,腹腔注射链脲佐菌素(STZ,40 mg.kg-1)复制糖尿病模型,采用RT-PCR方法,检测造模后6周肌肉组织PPAR-γ2 mRNA的表达。结果造模后6周,糖尿病模型组大鼠肌肉组织PPAR-γ2 mRNA的表达明显低于正常对照组(P<0.01)。结论 2型糖尿病伴有大鼠肌肉组织PPAR-γ2的表达水平下降。Objective To observe the expression of peroxisome proliferator activated receptor γ2 (PPAR-γ2) in the muscle tissue of rat model of type 2 diabetes, and to explore the pathogenesis of type 2 diabetes. Methods SD rats were randomly divided into control group(n=10) and model group(n=10). Rats in the model group were fed with high-fat diet for 2 months and then were injected intraperitoneally with STZ at 40 mg/kg. The expression PPAR-γ2 mRNA in the muscle tissue was detected by RT-PCR 6 weeks after modeling. Results PPAR-γ2 expression level in the muscle tissue of model rats 6 weeks after modeling was lower than in the normal control group, and there was sig- nificant difference(P 〈 0.01). Conclusion The decrease of PPAR-γ2 expression level in the muscle tissue occurs in type 2 diabetes rat model.
