益脑康胶囊对AS性AIS大鼠血清中内皮损伤相关因子的影响

摘　　要：目的研究益脑康胶囊对动脉粥样硬化(AS)性急性缺血中风(AIS)大鼠血液中一氧化氮(NO)、血管内皮生长因子(VEGF)、血小板衍生生长因子BB(PDGF-BB)等内皮损伤相关因子的影响,评价益脑康对AS性AIS的防治作用。方法采用一次性腹腔注射维生素D3联合高脂饲料喂养法复制AS大鼠模型。在此基础上采用内皮素-1诱导法建立AS+AIS复合模型。分别检测模型大鼠脑组织含水量,主动脉病变程度,血清NO、VEGF、PDGF-BB的水平。结果益脑康预防组、益脑康治疗组和立普妥阳性对照组大鼠脑组织含水量低于AS+AIS模型组(P<0.01)。益脑康预防组、立普妥治疗组血清NO水平高于AS+AIS模型组(P<0.01,P<0.05)。三个干预组血清VEGF、PDGF-BB水平低于AS+AIS模型组(P<0.01,P<0.05)。结论益脑康胶囊预防或治疗性用药能够提高血清NO水平,降低VEGF、PDGF-BB水平,可以减轻血管内皮损伤、纠正内皮功能紊乱、延缓动脉粥样硬化和抗血栓形成,可能对于动脉粥样硬化和急性缺血中风具有防治作用。Objective To evaluate the function of Yinaokang capsule preventing and treating atherosclerotic（AS） a- cute ischemic stroke（AIS）, it is studied that Yinaokang capsule effect on endothelial injury associated factors such as NO, VEGF, PDGF-BB in AS＋AIS rats＇ serum. Methods Given intraperitoueally injection of VD3 one time com- bined with high fat feed, AS model rats are copied. And on that basis by ET-1 inducement method, AS＋AIS model is established. Brain water content of rats, aortic lesion degree, and levels of NO, VEGF, PDGF-BB in serum are measured respectively and compared reciprocally. Results Rats＇ brain water content in three intervention groups are lower than those in AS＋AIS model group, and there were significant differences（P 〈 0.01 ）. NO levels of rats＇ serum in Yinaokang prevention group and Lipitor treatment group are higher than those in AS＋AIS model group, and there were significant differences（P 〈 0.01, P 〈 0.05）. VEGF, PDGF-BB levels of rats＇ serum in three intervention groups are lower than those in AS＋AIS model group, and there were significant differenees（P 〈 0.01, P 〈 0.05）. Con- clusions AS＋AIS rats intervened by Yinaokang capsule, their NO levels in the serum could be increased, VEGF, PDGF-BB content in the serum could be decreased. It can alleviate vascular endothelial injury, mend endothelial dysfunction and inhibit atherosclerosis as well as thrombosis. Hence, for Yinaokang capsule, there is preventive and curative function in treating atherosclerotic acute ischemic stroke.
