摘 要:目的建立同时测定檀香叶中牡荆苷与异牡荆苷含量测定方法,为其资源利用等提供依据。方法采用Agilent TC—C18色谱柱(4.6mm×250mm,5μm),流动相为甲醇-0.5%冰醋酸溶液(40:60),检测波长为338nm,柱温为30℃,流速为1mL·min^-1。结果牡荆苷的线性范围为7.625~488.0μg·mL^-1;高、中、低剂量回收率分别为96.38%、97.23%、97.96%,RSD为1.29%~1.83%。异牡荆苷的线性范围为8.047~515.01μg·mL^-1,高、中、低剂量回收率分别为97.81%、96.13%、97.98%,RSD为1.31%~1.81%。三批样品中牡荆苷与异牡荆苷的含量为1.13%~1.25%和1.20%~1.46%。结论檀香叶中牡荆苷与异牡荆苷含量较高,所建立的含量测定方法简便、准确,可用于檀香叶的质量控制。Objective To develop a HPLC method for simultaneous determination of vitexin and isovitexin in leaves of Santalum album L. Methods The separation was achieved on a C18 column(4.6 mm × 250 mm, 5 μm. Agilent, USA) with a mobile phase consisting of methanol-0.5 % acetic acid(40 : 60). The flow rate was 1 mL·min^-1. The column temperature was maintained at 30 ℃. The detection wavelength was at 338 nm. Results The linear range of vitexin was 7.625-488.0 μg·mL^-1. The recoveries of high-, middle- and low-concentration of vitexin were 96.38 %, 97.23 % and 97.96 % respectively, and the RSD was 1.29 %-1.83 %. The linear range of isovitexin was 8.047-515.0 μg·mL^-1. The recoveries of high-, middle- and low-concentration of isovitexin were 97.81%, 96.13 % and 97.98 % respec- tively, and the RSD was 1.31%-1.81%. The average content of three batches of samples was 1.13 %-1.25 % for vi- texin and 1.20 %-1.46 % for isovitexin. Conclusion The method is accurate and repeatable, which can be used for quality evaluation of vitexin and isovitexin of leaves of Santalum album L.
