Effect of Pyridoxamine and Telmisartan on Oxidative Stress of Rat Cardiac Myocytes Induced by Angiotensin Ⅱ

摘  要:目的探讨吡哆胺和替米沙坦对血管紧张素Ⅱ诱导的大鼠心肌细胞氧化应激的影响。方法大鼠心肌细胞培养并随机分为正常对照组(C组),血管紧张素Ⅱ组(1×10-6mol/L,AngⅡ组),吡哆胺组[吡哆胺(10 mmol/L)+AngⅡ(1×10-6mol/L),P组],替米沙坦组[替米沙坦(1×10-6mol/L)+AngⅡ(1×10-6mol/L),T组],联合治疗组[替米沙坦(1×10-6mol/L)+吡哆胺(10 mmol/L)+AngⅡ(1×10-6mol/L),TP组]。流式细胞仪检测细胞上清液中的活性氧簇(ROS)浓度,硫代巴比妥酸法(TBA)测定丙二醛(MDA)浓度,黄嘌呤氧化酶法测定超氧化物歧化酶(SOD)活力,荧光实时定量PCR方法检测心肌细胞所表达的晚期糖基化终产物受体(RAGE)mRNA表达量。结果与AngⅡ组比较,T组、P组及TP组MDA、ROS浓度均显著降低、SOD活力显著升高(P<0.05);与T组比较,P组、TP组的ROS浓度明显降低、SOD活力明显升高(P<0.05);与AngⅡ组比较,T组、P组和TP组RAGE mRNA表达量明显降低(P<0.01);与T组、P组比较,TP组的RAGE mRNA表达进一步降低(P<0.05)。结论吡哆胺和替米沙坦协同下调大鼠心肌细胞RAGE mRNA的表达,二者均可改善氧化应激,但吡哆胺较替米沙坦强。Objective To investigate the protective effect of telmisartan and pyridoxamine on oxidative stress in cultured rat cardiac myocytes. Methods Cultured cardiac myocytes were divided into control group (C), angiotensin Ⅱ (1× 10^-6 mol/L) group (Ang Ⅱ ), pyri- doxamine group (pyridoxamine 10 mmol/L,P),telmisartan group (telmisartan 1×10^-6 mol/L+ Ang Ⅱ 1×10^-6 mol/L,T),and T + P + an- giotensin H group (telmisaaan 1×10^-6 mol/L + pyridoxamine 10 mmol/L+Ang Ⅱ 1×10^-6 mol/L,TP). Reactive oxygen species (ROS)genera- tion in cellular supematant was detected by flow cytometry. The maleic dialdehyde (MDA) was detected by thiobarbituric acid (TBA). Xan- thine oxidase was performed to determine super oxide dismutase (SOD) in cellular supematant. Real-time quantitative PCR was used to mea- sure the mRNA expression level of receptor for advanced glycation end-products (RAGE). Results Compared with the Ang Ⅱ group, the level of ROS and MDA was reduced,while SOD activity was increased in T,P and TP groups (P 〈 0.05). Compared with the T group,the level of ROS was reduced,while SOD activity was increased in P and TP groups (P 〈 0.05). Compared with the Ang H group,the mRNA expression level of RAGE was decreased significantly in T,P and TP groups (P 〈 0.01 ). Compared with T and P groups,mRNA expression in RAGE was much lower in TP group (P 〈 0.05 ). Conclusion There is a synergistic effect on reducing rat cardiac myocytes RAGE by combined use of telmisartan and pyfidoxamine. Pyridoxamine and telmisartan can improve oxidative stress in rat cardiac myocytes and pyri- doxamine is more effective than telmisartan.
