摘  要:目的探讨雷公藤多甙致小鼠生殖系统损害及雌孕激素替代对生殖系统保护的机制。方法选取30只成年雌性KM小鼠,随机分成雷公藤多甙组、雌孕激素干预组(干预组)、正常对照组。雷公藤多甙组每日给予雷公藤多甙灌胃,干预组每日给予雷公藤多甙灌胃和雌孕激素序贯性干预治疗,对照组每日给予等量生理盐水灌胃,为期12周。采用ELISA分别测定3组小鼠血清中性激素水平的变化,各组卵巢组织进行HE染色并观察卵巢组织的变化,免疫组织化学染色法检测卵巢组织中凋亡相关蛋白Fas和FasL的表达水平。结果与正常对照组相比,雷公藤多甙组小鼠血清黄体生成素和卵泡刺激素水平明显升高,雌激素分泌明显下降,差异有统计学意义(P<0.01);卵巢组织受损明显,卵泡较小,各级卵泡均明显减少,且闭锁卵泡增多。干预组小鼠血清性激素水平及卵巢组织较正常对照组无明显变化。凋亡相关蛋白Fas和FasL的表达在雷公藤多甙组明显高于干预组和正常对照组(P<0.05),而干预组和正常对照组比较Fas和FasL表达的差异无统计学意义(P>0.05)。结论雷公藤多甙可致雌性小鼠生殖功能损害,采用雌孕激素替代治疗对小鼠卵巢的功能有较好的保护作用,可促进雷公藤多甙所致受损卵巢功能的恢复,其保护机制可能与雌孕激素抑制了卵巢组织Fas和FasL蛋白的表达有关。Objective To investigate tripterygium wilfordii polyglycosidium (TWP)-induced injury of female mouse genital system,and ex- plore the protective mechanism of estrogen-progestogen replacement therapy on genital system. Methods A total of 30 female KM mice were randomly divided into three groups:TWP group,intervention group,and control group. The process lasted for 12 weeks. The levels of serom gonadal hormone were detected by ELISA. The histological changes of the mouse ovary were observed after HE staining. The expres- sion levels of Fas and FasL protein in ovary were detected by immunohistochemical staining technique. Results Compared with the control group, the levels of LH and FSH secreted by pituitary were higher and the level of E2 secreted by ovary was lower in TWP group (P 〈 0.01 ). There was no significant difference of these three hormones levels between intervention group and control groups (P 〉 0.05). In TWP group, ovarian tissues were significantly injured with obvious decreasing of follicles and more clausura of these follicles. Compared with control group,intervention group could restrain ovary pathological changes induced by TWt'. Fas and FasL expression in mouse ovary cells were higher in TWP group compared with control group;and there was no significant difference of Fas and FasL expression between control group and intervention group (P 〉 0.05). Conclusion TWP can induce genital system injury of mouse, hormone replacement therapy has a good protective effect on mouse ovarian function and can promote the restoration of impaired ovar/an function. The mechanism may be partly relat- ed to inhibition of Fas and FasL expression in ovarian tissues.
