摘 要:目的:研究附子、黄芩和穿心莲水提物对四氢吡啶离子(MPP+)诱导SH-SY5Y细胞凋亡的影响。方法:体外培养SH-SY5Y细胞,建立MPP+致SH-SY5Y细胞凋亡的模型,同时给予附子、黄芩和穿心莲水提物干预。应用MTT比色法检测细胞活力,流式细胞仪检测细胞凋亡率。结果:0.5mmol·L-1MPP+作用于SH-SY5Y细胞60h可诱导细胞凋亡,MTT值降低至(30.43±2.69)%,Hoechst33258染色可见明显的颗粒状和固缩状荧光,凋亡率增加至(40.52±1.68)%。附子水提物能明显改善以上凋亡特性,显著升高MTT值,降低凋亡率;黄芩和穿心莲水提物则对MPP+诱导的SH-SY5Y细胞凋亡几无影响。结论:附子对MPP+诱导的SH-SY5Y细胞凋亡有显著保护作用,而黄芩、穿心莲却无此保护作用,中药药性不同,其抗凋亡作用亦不同。OBJECTIVE: To study the effects of water extracts of Aconitum carmichaeli, Scutellaria baiealensis and Androgra- phis paniculata on SH-SYSY cells apoptosis induced by MPP+. METHODS: SH-SYSY cells were cultred in vitro and MPP+ in- duced SH-SYSY cells apoptotic model was established and treated with water extracts ofA. carmichaeli, S. baicalensis and A. panic- ulata. The cell activity was measured by MTT test, and the cell apoptosis rate was detected by flow cytometer. RESULTS: MTT de- creased remarkably to (30.43 ± 2.69) %, the apoptosis of SH-SY5Y cells was induced by 0.5 mmol.L-1 MPP+ for 60 h and were positively stained with fluorochrome Hoechst 33258, presenting the apoptosis rate increased to (40.52 ± 1.68)%. However, water extracts of A. carmichaeli obviously promoted apoptosis property, improved MTT value and reduced apoptosis, and water extracts of S. baicalensis and A. paniculata almost had no effect on the apoptosis of SH-SYSY cells. CONCLUSION:A. carmichaeli has markedly protective effects on SH-SY5Y cells apoptosis induced by MPP+, but S. baicalensis and A. paniculata shows no protective effects. As the different natures of TCM, the anti-apoptotic effects of TCM are different neither.
