摘 要:目的优选武当山野生半枝莲多糖提取工艺,并对其多糖含量进行测定。方法以多糖得率为考察指标,在单因素试验的基础上选定温度、时间、提取次数和固液比4因素进行L9(34)正交试验;蒽酮-硫酸分光光度法测定半枝莲中多糖含量。结果在实验设计水平范围内,提取多糖最佳工艺条件为温度80℃、提取3次、时间4h、固液比1:25。该工艺条件下半枝莲多糖得率为7.60%,RSD=2.12%(n=3)。半枝莲中多糖含量为4.31%,RSD=1.67%(n=5),加样回收率平均值为(99.76±1.43)%,RSD=1.35%(n=5)。结论该优化工艺可用于半枝莲多糖的提取,得率稳定,提取率高;武当山野生半枝莲多糖含量较高,有进一步开发利用价值。OBJECTIVE To optimize the extracting conditions of polysaccharide from Scutellaria barbata D.Don in Wudang mountains and establish a suitable method for its content determination. METHODS Based on the qualitative yield of polysac- charide extraction, a orthogonal test was employed to evaluate the effect of four factors including extracting temperature, ex- tracting time, the extraction times and the ratio of material to liquid. Selected amount of flavonoids as a marker according to L9(3^4) scheme. The colorimetry of anthronesulfuric acid method was applied for the content determination of pelysaccharide. RESULTS The optimum extraction was described as follows: the material to water ratio was 1 : 25, and the Seutellaria barbata D.Don was extracted 3 times at 80℃ with 4 hours for each time. In this conditions, the polysaccharide content was 7.60%, RSD = 2.12% (n = 3). The content of polysaccharide in Scutellaria barbata D.Don was 4.31%, RSD = 1.67% (n = 5). The average recovery was (99.76 ± 1.43) %, RSD = 1.35%. CONCLUSION The proposed method has been applied stably and reliably to the extraction of polysaccharide from Seutellaria barbata D.Don. Scutellaria barbara D.Don in Wudang mountains has high polysaceharide for further development and utilization.
