摘 要:目的研究HIV-1 DNA疫苗的免疫效价及不同剂量HIV-1 DNA疫苗对小鼠诱导免疫反应的影响。方法将80只BALB/c小鼠随机分为8组,每组10只,分别为对照组(pDRVI1.0空载体,100μg)、DNA疫苗pENV 3.125、6.25、12.5、25、50、100和200μg组,经小鼠胫骨前肌注射,100μl/只,每隔2周免疫1次,共3次。采用ELISA法检测小鼠血清抗体反应,并计算抗体阳转率;ELISPOT法检测小鼠T细胞免疫反应,并计算阳转率。结果高剂量组抗体反应仅25μg与100μg组之间差异有统计学意义(P<0.05),其他高剂量组间及低剂量各组间差异均无统计学意义(P>0.05),而各疫苗组抗体阳转率总体之间差异有统计学意义(P<0.01)。高剂量组细胞免疫反应强度之间差异无统计学意义(P>0.05),且比低剂量组更趋稳定。DNA疫苗诱导的抗体和细胞免疫反应均随免疫剂量的增加而增加,且呈正相关。当疫苗剂量达到100μg时,均达最高反应强度。结论 HIV-1 DNA疫苗25μg免疫剂量对BALB/c小鼠可能是适当的免疫剂量。在选择临床或临床前试验免疫剂量时,使用统计学无差异的低剂量可能免疫效果更好。Objective To investigate the immunological potency of HIV-1 DNA vaccine as well as the immune response induced by vaccine at various dosages.Methods Eighty BALB / c mice were randomly divided into 8 groups,10 for each.The mice in seven test groups were injected i.m.with DNA vaccine pENV at dosages of 3.125,6.25,12.5,25,50,100 and 200 μg respectively,while those in control group with 100 μg empty vector pDRVI1.0,for 3 times at intervals of 2 weeks.The serum antibodies of mice were determined by ELISA,based on which the antibody positive conversation rate was calculated.The T cell responses of mice in various groups were determined by ELISPOT,based on which the response rate was calculated.Results The antibody response levels showed significant difference in the mice immunized with 25 and 100 μg DNA vaccine pENV(P 〈 0.05),while showed no significant difference in other high dosage(not less than 25 μg)groups,or in low dosage(less than 25 μg)groups(P 〉 0.05).Generally significant differences were observed in antibody positive rates in various test groups(P 〈 0.05).The cellular immune response levels showed no significant difference in high dosage groups(P 〉 0.05),which were more stable than those in low dosage groups.Both the antibody and cellular response levels were positively related to the dosage of vaccine,of which those induced by 100 μg vaccine reached peak values.Conclusion The HIV-1 DNA vaccine at a dosage of 25 μg might be suitable for BALB/c mice.However,low dosages without significant difference might be more suitable for pre-clinical test or clinical trial.
