摘  要:目的探讨红豆杉醇提物作为H3N2流感病毒灭活疫苗佐剂对小鼠的免疫原性及安全性。方法将ICR小鼠随机分为5组,每组7只,醇提红豆杉佐剂组:含血凝素1.2μg,红豆杉醇提物0.2 mg;水提红豆杉佐剂组:含血凝素1.2μg,红豆杉水提物0.2 mg[3];铝佐剂对照组:含氢氧化铝0.2 mg,血凝素1.2μg;疫苗对照组:含血凝素1.2μg;空白对照组:注射生理盐水0.2 ml。分别于0、21 d经腿部肌肉注射免疫2次,每次0.2 ml/只,初免后每隔7 d采血,收集血清,检测血凝抑制(HI)抗体效价,于初免后1、21、70 d观察各组小鼠的体重变化,并测定红豆杉醇提物的小鼠半数致死量(LD50)。结果醇提红豆杉佐剂组各时间点的小鼠血清HI抗体效价均高于疫苗对照组,在28 d时,含佐剂疫苗组的抗体效价均高于单独疫苗组,醇提红豆杉佐剂组的抗体产生先于铝佐剂对照组;各组小鼠体重增长差异无统计学意义(P>0.05);红豆杉醇提物的LD50值为577.93 mg/kg。结论红豆杉醇提物作为流感疫苗佐剂具有良好的安全性及有效性,有望成为流感灭活疫苗的新型免疫佐剂。Objective To investigate the immunogenicity and safety of alcohol extract of Taxus madia as adjuvant of influenza H3N2 virus vaccine in mice.Methods ICR mice were divided into five groups randomly,and injected i.m.twice with influenza H3N2 virus vaccine containing 1.2 μg of hemagglutinin(HA)and 0.2 mg of alcohol extract of Taxus madiaf,the vaccine containing 1.2 μg of HA and 0.2 mg of water extract of Taxus madia,the vaccine containing 1.2 μg of HA and 0.2 mg of aluminum hydroxide,adjuvant-free vaccine containing 1.2 μg of HA,and physiological saline,respectively,on days 0 and 21,0.2 ml for each.Serum samples were collected every 7 d after the first injection and determined for HI titer.The mice in various groups were observed for bodyweights on days 1,21 and 70,while the LD50 of alcohol extract of Taxus madia was determined.Results The HI antibody titer induced by the vaccine containing alcohol extract of Taxus madia was significantly higher than those induced by adjuvant-free vaccine.On day 28,all the antibody titers induced by adjuvant-containing vaccines were higher than those by adjuvantfree vaccine.However,the antibody appeared early in the mice immunized with the vaccine containing alcohol extract of Taxus madia as adjuvant as compared with those containing aluminum adjuvant.No significant differences were observed in the increases of bodyweights of mice in various groups(P 〉 0.05).The LD50 of alcohol extract of Taxus madia was determined as 577.93 mg / kg.Conclusion The alcohol extract of Taxus madia showed high safety and effectiveness,which might be used as a novel immune adjuvant of inactivated influenza vaccine.
