摘  要:目的探讨黄芪葛根汤(黄芪、葛根等)对糖尿病心肌氧化应激、核转录因子(NF-κB)mRNA表达的影响。方法采用高脂饲料喂养和链脲佐菌素腹腔注射(30 mg/kg)的方法建立2型糖尿病大鼠心肌病模型,连续给药8周,观察黄芪葛根汤对大鼠一般体征和血糖、血脂的影响,生化法检测心肌组织超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)的活性变化,RT-PCR方法检测NF-κB mRNA表达。结果黄芪葛根汤可改善糖尿病心肌病大鼠多饮、多尿、多食的症状,明显降低大鼠空腹血糖、血脂,显著抑制MDA的产生,防止SOD、GSH-Px活性下降,明显抑制心肌NF-κB mRNA的过表达。结论黄芪葛根汤可能通过减轻氧化应激反应,下调糖尿病大鼠心肌组织中NF-κB的表达水平实现对糖尿病心脏的保护作用。AIM To study the effects of Huangqi Gegen Decoction (Astragali Radix, Puerariae lobatae Radix, etc. ) on oxidative stress and expression of NF-KB in diabetic cardiomyopathy rats. METHODS Rats receiving high fat diet with STZ (ip, 30 mg/kg) were modeled into Type 2 diabetic cardiomyopathy rats with eight consecutive weeks' administration. The effects of Huangqi Gegen Decoction were observed on general signs, blood glucose and blood lipids in rats. SOD, MDA and GSH-Px activities of myocardial tissue were detected by biochemical assay and NF-KB mRNA expression was detected by RT-PCR. RESULTS Huangqi Gegen Decoction obviously improved polydipsia, polyuria, polyphagia symptoms of diabetic rats, significantly reduced fasting blood glucose, blood lipids, significantly inhibited the production of MDA, prevented the decline in the activities of SOD, GSHPx, and obviously inhibited over-expression of NF-kB mRNA. CONCLUSION Huangqi Gegen Decoction may protect heart of diabetes by reducing oxidative stress and lowering level of NF-KB expression in the heart tissue.
