摘 要:目的对重楼中的成分进行分离鉴定,结合药理模型筛选具有抑制血管生长活性的成分。方法采用乙酸乙酯提取和硅胶柱层析法进行提取分离;利用1HNMR、13CNMR、13CNMR-APT、MS、IR、UV确定其化学结构,并且应用鸡胚绒毛尿囊膜模型进行活性筛选。于鸡胚发育的第7天,将加药后的明胶海绵载体加于鸡胚绒毛尿囊膜上,孵育2 d后,固定鸡胚绒毛尿囊膜,取出观察。结果从重楼中分离得到了4个甾体皂苷类成分,分别为重楼皂苷Ⅰ、重楼皂苷Ⅱ、重楼皂苷Ⅴ和纤细薯蓣皂苷。通过活性筛选发现重楼皂苷Ⅰ、重楼皂苷Ⅱ、纤细薯蓣皂苷3组鸡胚尿囊膜的小血管数目均小于空白组,其中重楼皂苷I经统计分析P<0.05具有显著性差异。结论重楼皂苷Ⅰ具有抑制血管生长的活性。AIM To isolate and identify steroidal saponins from Paridis Rhizoma and to investigate the effects of steroidal saponins on the angiogenesis of chick embryo chorioallantoic membranes. METHOD Paridis Rhizoma was extracted by ethyl acetate and separated by chromatography on gel silica. The structures were determined by 1 HNMR, ~3 CNMR, 13 CNMR-APT, MS, IR and UV. Steroidal saponins were implanted on the top of chick embryo chorioallantoic membranes when chick embryos developed up to the seventh day. The numbers of vessels were counted. RESULTS Four compounds were obtained and elucidated as polyphyllin I , II, V and gracillin. The numbers of chick embryo chorioallantoic membranes small blood vessels induced by polyphyllin I , II and gracillin were less than those of the control group. There was a significant difference in the blood vessels numbers of polyphyllin I group (P 〈 0. 05 ). CONCLUSION Polyphyllin I has the activity of the angiogenesis of chick embryo ehorioallantoic membranes.
