摘  要:目的:观察头穴丛刺对慢性脑缺血大鼠学习记忆能力和HIF-1α及其靶基因HO-1表达的影响。方法:采用改良的双侧颈总动脉永久结扎术(2VO)制造慢性脑缺血模型,将大鼠随机分为假手术组及手术组,手术组包括尼莫地平组、头穴丛刺组、模型组,用Morris水迷宫检测大鼠学习记忆能力,HE染色及免疫组化观察额叶皮层、海马病理变化和HIF-1α及其靶基因HO-1的表达。结果:治疗4周后头穴丛刺组和尼莫地平组与模型组相比,逃避潜伏期明显缩短(P<0.05),平台所在象限游泳路程明显延长(P<0.05),头穴丛刺组和尼莫地平组中HIF-1α及HO-1阳性表达均较模型组明显增多(P<0.05),假手术组几乎不表达。结论:头穴丛刺可能通过影响HIF-1α及其靶基因HO-1的表达达到神经保护作用,减轻神经细胞损伤,从而改善慢性脑缺血大鼠学习记忆能力。Objective:To observing the effects of scalp clustery acupuncture to learning and memory ability and the expression of hypoxia inducible factor - 1 alpha and its target gene hemeoxygenase 1 of chronic cerebral is- chemic rats. Methods:Sarti's 2VO was used to make the model of chronic cerebral ischemic rats, and divided the rats into fake operation group and operation group. The operation group includes nimodipine subgroup, scalp clustery acupuncture subgroup and model subgroup. The morris water maze was used to detect learning and memory ability of rats. HE and immunobistochemistry were used to detect the ability of learning and memo- ry and the expression of hypoxia inducible factor - 1 alpha its target gene hemeoxygenase 1. Results:Statistics showed that the escaped incubation period is obviously shortened ( P 〈 0.05 ), swimming path belong to the plat- form quadrant is obviously extened ( P 〈 0.05 ), and the positive expression of hypoxia inducible factor - 1 alpha its target gene hemeoxygenase 1 were increased significantly ( P 〈 0.05 ) in the nimodipine subgroup and scalp clustery acupuncture subgroup after 4 weeks treatment. And it almost can not be expressed in the fake operation group. Conclusion:Scalp clustery acupuncture can improve the ability of learning and memory of chronic cere- bral ischemic rats a lot. The mechanism probably related to the influence of hypoxia inducible factor - 1 alpha and its target gene hemeoxygenase 1 expression to protect nerve after scalp clustery acupuncture.
