摘  要:目的:通过对代谢综合征大鼠主动脉核转录因子及肿瘤坏死因子的检测,探讨黄连温胆汤对该指标的影响及其机制。方法:将8周龄雄性SD大鼠分组采用膳食饲料喂养,建立MS大鼠模型后将MS大鼠随机分为模型组,格华止组,黄连温胆汤高、低剂量组,灌胃4周后,检测体质量、血压、血糖、血清胰岛素、血脂、TNF-α,计算lee's指数和胰岛素敏感指数,测定大鼠主动脉NF-κB的表达。结果:与空白组比较,MS组大鼠体质量、空腹血糖、血脂等差异均有统计学意义(P<0.05)。黄连温胆汤高剂量组体质量、血脂、Lee's指数、HOMA-IR、TNF-α,与MS组大鼠比较均有统计学意义(P<0.05),并且三组大鼠主动脉NF-κB表达均有统计学意义(P<0.05)。结论:黄连温胆汤MS大鼠各组分有治疗作用,并且可以降低MS大鼠血清TNF-α,改善主动脉NF-κB的表达,其机制可能与核转录因子分子通路有关。Objective:To explore the effective and mechanism of the treatment of HuangLianWenDan decoction for MS by detecting the lever of NF - KB and TNF - a in serum of MS rats. Methods : The 8 - week - old male SD rats were fed with high salt, high lipid and high glucose diet to build model of MS and the successful model rats were randomly divided into 4 groups: the model group, metformin group, huanglianwendan decoction high dose group,and low dose group. After gavaging the corresponding drug to rats 4 weeks, detect the weight, blood pressure, blood glucose, serum insulin, blo- od lipids, TNF-a and calculate the leeg index, HOMA - IR, and measure the expression of NF - KB. Results : Compared with normal group, the weight, fast blood glucose, blood lipids of model group rats were significantly different (P 〈 0.05 ). Contrasting to model group, the weight, blood lipid, lee's index, HOMA - IR, TNF - a of MS rats in high dose group had significantly difference ( P 〈0. 05 ) group. And the expression of NF - KB in three groups had significantly difference ( P 〈 0. 05 ). Conclusions : HuanglianWendan Decoction has treatment on each aspart of MS rats. It can reduce the serum TNF -a concentration and improve aortic NF - KB expression. The mechisim of the process may be re- lated with NF - KB molecular pathways.
