摘 要:目的:观察中药复方抗痫灵对戊四氮点燃癫痫大鼠模型脑电图及海马NMDAR1表达的影响。方法:Wistar雄性大鼠60只,体质量180~220g,随机分成空白组、模型组、抗痫灵高、中、低剂量组、丙戊酸钠(VPA)组各10只。点燃后各组大鼠描记脑电图,低温条件下迅速断头取脑,用免疫印迹(Westernblot)方法观察各实验组大鼠海马的NMDAR1活性变化。结果:抗痫灵组、丙戊酸钠组、模型组阳性细胞表达与空白组相比有显著性差异(P<0.05),抗痫灵组显著低于模型组(P<0.01)。结论:抗痫灵对戊四氮点燃大鼠癫痫发作有显著对抗作用,其作用机制可能与其降低模型大鼠海马NMDAR1活性有关。Objective : To observe the effect of Kangxianling on the EEG and the expression of NMDAR1 in hip- pocampus of the PTZ - induced epilepsy model of rats. Methods : PTZ was used to evoke chronic epilepsy model. The Wister rats were randomly divided into control group, PTZ group ; low - dose Kangxiangling group; moderate- dose Kangxianling group;high -dose Kangxianling group;VPA group. Ethology changes and electro encephalography were observed after epileptic seizure, expressions of NMDAR1 were investigated with Western blot method. Results:The group of Kang xianling, VPA group and PTZ group were significantly different from the control group ( P 〈 0. 05 ). The group of Kangxianling was siginificantly different from the model group ( P 〈 0. 01 ). Conclusion : Kangxianling has an anti - epileptic seizure resistant function on PTZ - induced epilepsy-model of rats. Its functional mechanism may be to react by reducing the content of NMDAR1 in the hippocampus.
