摘 要:目的:观察桦菌芝多糖对小鼠移植型肉瘤(S180)的抑制作用及对瘤组织p53蛋白表达的影响。方法:建立S180小鼠动物模型,取瘤组织称其质量,计算抑瘤率;用链霉菌抗生素蛋白-过氧化酶免疫组化法来测定瘤组织中p53蛋白含量,通过多媒体图像分析系统分析实验结果。结果:桦菌芝多糖高、中剂量组抑瘤率分别为41.84%、41.13%,桦菌芝多糖高、中剂量组S180瘤组织突变型p53蛋白表达减少,与模型组比较差异显著(P<0.05)。结论:桦菌芝多糖对小鼠肉瘤S180有抑制作用,可下调小鼠S180瘤组织突变型p53基因表达。Objective: To study the effect on p53 protein expression in S180 tumor tissue with Pyropolyporus Fomentaries polysaccharide (PFPS) and inhibition of sarcoma - S180 cell proliferation of mice. Methods: The tumor weight was measured to calculate the tumor inhibition rate. Immunohistochemistry method was used to detect p53 protein in tumor tissue treated with PFPS. Results: The PFPS treatment at 100 and 50mg/kg re- sulted in tumor growth inhibition rates of 41.84% and 41.13%. Immunohistochemistry analysis demonstrated the expression level of mutatant p53 decreased. There was a significant difference compared with the negative control group(P 〈0. 05). Conclusion:The PFPS has an inhibitory effect against S180 sarcoma in vivo and can decrease the expression level of mutatant p53.
