摘 要:为寻求评价发酵产氢活性污泥产氢效能的合理指标,以氯化三苯基四氮唑(TTC)检测法为基础,优化了TTC-比脱氢酶活性检测方法,并分析了发酵产氢活性污泥的TTC-比脱氢酶活性与比产氢速率的相关性。结果表明,发酵产氢活性污泥TTC-比脱氢酶活性的适宜检测方法及条件为:向10 mL离心管中依次加入污泥样品1 ml(8.47~26.39 g MLVSS.L-1)、17.8mmoL.L-1的TTC溶液1 ml、pH 8.0的Tris-HCl溶液2.5 ml和0.36%的Na2SO3溶液0.5mL,35℃暗处振荡反应20 min;加98%浓硫酸2 ml终止反应,以无水乙醇5 ml萃取反应产物三苯基甲,475 nm测定萃取液吸光度。发酵产氢活性污泥的TTC-比脱氢酶活性,与比产氢速率的相关系数R〉0.8,是反映发酵产氢活性污泥产氢活性的一个优良指标。In order to find rational index of evaluating efficiency of fermentative hydrogen production activated sludge hydrogen production,the paper optimizes triphenyl tetrazolium chloride-specific dehydrogenase activity detection method based on triphenyl tetrazolium chloride detection method and analyzes the relationship between triphenyl tetrazolium chloride-specific dehydrogenase activity of fermentative hydrogen production activated sludge and hydrogen productivity.The result shows adaptive detection method and condition of triphenyl tetrazolium chloride-specific dehydrogenase activity of fermentative hydrogen production activated sludge are putting 1mL sludge sample(8.47~26.39 g MLVSS·L-1),1mL TTC solution which concentration is 17.8 mmoL·L-1,2.5mL Tris-HCL solution which PH is 8.0 and 0.5mL Na2SO3 which concentration is 0.36% into 10 mL centrifuge tube,oscillating reaction in 20min in 35℃ under dark;termination reaction adding 2 mL concentrated sulfuric acid which concentration is 98%,extracts reaction product triphenylformazan with 5 mL ethanol,measures extract liquid absorbance at 475nm.The results show triphenyl tetrazolium chloride-specific dehydrogenase activity of fermentative hydrogen production activated sludge and hydrogen productivity correlation coefficient R is more than 0.8 and the coefficient is a good index to reflect fermentative hydrogen production activated sludge hydrogen activity.
