摘 要:目的构建叶酸(folic acid,FA)分子靶向磁性纳米载体,表征其基本理化性质,分析与基因的结合能力。方法将化学共沉淀法制备的醛基化海藻酸钠(Alg)改性的Fe3O4纳米与酰胺化反应制备的聚乙二醇(PEG)-聚乙烯亚胺(PEI)-FA化合物充分混合形成PEG-PE(I-FA)-Alg-Fe3O4纳米,使用透射电镜、激光粒度检测仪、磁强计、紫外分光光谱等检测该纳米形态、粒径及Zeta电位、磁饱和强度、FA成分等;采用琼脂糖凝胶电泳分析该纳米结合基因能力。结果该纳米分散均匀,平均水动力学粒径197.8 nm,Zeta电位+12.6 mV。具有超顺磁性。紫外分光光谱检测显示成功偶联FA分子。凝胶电泳结果显示纳米粒能有效结合质粒。结论该纳米颗粒能够携带基因,可用于磁靶向及分子靶向治疗实验。【Objective】 To construct FA targeted magnetic nanoparticles gene vector,characterize its basic physical and chemical properties,and analyze its capacity of binding gene.【Methods】 Aldehyde sodium alginates modified Fe3O4 magnetic nanoparticles prepared by a chemical co-precipitation method and a PEG-PEI-FA chemical compound prepared through amidation reaction are fully mixed to form PEG-PEI(-FA)-Alg-Fe3O4 nanoparticles,the nanoparticles such as morphology,particle size,zeta potential,magnetic saturation intensity,FA structure composition,etc are detected by using transmission electron microscopy,laser particle detector,magnetometer,UV spectra and the like;And its capacity of binding gene is analyzed by using agarose gel electrophoresis.【Results】 The nanoparticles is in uniform dispersion,its average hydrodynamic particle size is 197.8 nm,Zeta potential is +12.6 mV,UV spectra detection shows the successful coupling on FA,agarose gel electrophoresis shows that nanoparticles can be connected with plasmid effectively.【Conclusion】 The nanoparticles can carry gene and be used for magnetic targeting and molecular targeted therapy experiment.
