摘 要:目的:建立双山颗粒中绿原酸、芦丁和槲皮苷3种成分的HPLC含量测定方法。方法:色谱柱采用Diamonsil C18(2)柱(250 mm×4.6 mm,5μm);流动相为乙腈-0.2%磷酸溶液,梯度洗脱;检测波长为355 nm;柱温为25℃;流速为1.0 mL.min-1。结果:绿原酸、芦丁和槲皮苷的线性范围分别为0.106~0.636μg(r=0.999 7),0.035 3~0.211μg(r=0.999 4)和0.030 4~0.243μg(r=0.999 5);3成分的平均回收率在99.0%~100.1%。结论:该法简便、准确、重复性好,可用于双山颗粒剂的质量控制。Objective: To develop an HPLC method for determination of the three active components ( chlorogenic acid, rutin and quercitrin) in Shuangshan granules. Methods : HPLC analysis was performed at 25 ℃ on a Diamonsil C18 column (250 mm×4.6 mm, 5 μm) e]uted with a mobile phase consisting of acetonitrile-0.2% glacial acetic acid in gradient elution at a flow rate of 1.0 mL· min^(-1) The UV detection was set at 355 nm. Results: A satisfactory linear relation existed between the peak area and injected amount in range of 0. 106 -0. 636 μg for chlorogenic acid ( r = 0. 999 7), 0. 035 3 - 0.211 μg for rutin ( r = 0. 999 4), and 0. 030 4 - 0. 243 2 μg for quercitrin (r = 0. 999 5). The average recoveries of the three components were from 99.0% to 100.1%. Conclusion: The method is sensitive, specific and accurate with a good repeatability, and can be used for the quality control of Shuangshan granules.
