摘 要:目的:研究不同剂量附子长期使用后心力衰竭大鼠心室重构、神经内分泌、血流动力学水平的变化。方法:结扎90只SD大鼠冠状动脉前降支,4周后形成心衰模型,随机分为模型组、假手术组、卡托普利阳性对照组、附子高剂量组(9 g.kg-1)、附子低剂量组(4.5 g.kg-1),造模成功后即开始给药,观察不同剂量的附子水煎液ig 4周后对慢性心力衰竭大鼠的血流动力学、神经内分泌水平和心室重构等指标的影响,并与卡托普利相对照。结果:90只SD雄性大鼠经造模及血流动力学检测后共有50只完成实验,附子高剂量组左室质量指数(LVMI)比各组都有明显增加,血流动力学指标与其他各组相比下降(P<0.05),低剂量组与卡托普利组上述2项指标与模型组无差别,血清血管紧张素Ⅱ(AngⅡ)附子高剂量组高于假手术组、卡托普利、附子低剂量组(P<0.05)。结论:高剂量使用附子后心衰大鼠血流动力学和神经内分泌水平恶化,心室重构加重;低剂量附子与卡托普利作用类似,能够改善神经内分泌紊乱。Objective: To investigate different doses of 'Aconiti Lateralis Radix P.raeparatat' in regulating ventricular remodeling, neuroendocrine, hemodynamic in chronic heart failure (CHF) rats. Method: A rat model of CHF was established by ligating left anterior descending coronary artery way, ninety Sprague-Dawley (SD) rats were randomly assigned into sham-operation group, model group, captopril-control group, decoction of Aconiti Lateralis Radix Praeparata low (4.5 g·kg^-1 ), high (9 g·kg^-1 ) dose group, drug intervention was started on 28^th day after operation, corresponding drugs was given to each treatment group by garage for 4 weeks. Meanwhile, the same volume of physiological saline was given to the model group and the sham-operation group by gavage once a day for 4 weeks. The hemodynamic, neuroendocrine, ventricular remodeling data were detect by normal way. Result: Fifty rats were survived after operation and observation of hemodynamic data, left ventricular remodelin'g and angiotension Ⅱ in high dose group were greatly deteriorated than model group, captopril group and low dose group (P 〈 0.05). Data of hemodynamic were significantly decreased in high dose group (P 〈 0.05). Angiotension II in captopril and low dose group were lower than model group (P 〈 0.05). Conclusion : High dose usage of Aconiti Lateralis Radix Praeparata may harm hemodynamic, neuoendocrine and ventricular remodeling of CHF rats. Like captpril, low dose usage of ' prepared aconite root' can moderate neuroendocrine of CHF rats.
