丁香降气方对反流性食管炎大鼠IL-8及其受体的影响

摘　　要：目的观察丁香降气方对反流性食管炎模型大鼠食药组织IL-8及其受体的影响。方法采用下食管括约肌切开术加十二指肠半结扎术建立混合反流性食管炎大鼠模型。52只健康SD大鼠随机分为正常组、模型组、中药组和西药组,每组13只,给予相应干预措施。在光镜下观察食管组织的形态学改变,并应用免疫组化和放射免疫的方法检测IL-8和CXCR-1在反流性食管炎大鼠食管组织的表达。结果模型组病理积分明显高于正常组、中药组、西药组,差异有统计学意义(P<0.05)。与正常组比较,模型组IL-8含量及IL-8蛋白表达升高,差异有统计学意义(P<0.05,P<0.01);与模型组比较,中药组、西药组IL-8含量及IL-8蛋白表达降低,差异有统计学意义(P<0.01)。各组大鼠食管组织CXCR-1蛋白表达差异均无统计学意义(P>0.05)。结论丁香降气方能改善RE大鼠食管黏膜的病理状况,其作用机制可能与下调模型大鼠食管黏膜炎性细胞因子IL-8的含量、减轻食管炎症有关。Objective To observe the clinical efficacy and mechanism of ＂Dingxiang Jiangqi Decoction＂ on reflux esophagitis. Methods The models were established by esophageal sphincter dissection and duodenal half ligation. Fifty-two SD rats were randomized into rmrmal group, model group, Chinese medicine group and western medicine group, 13 rats in each group were treated by CO^Tesponding interventions. The morphological changes of esophageal tissues were detected by light microscope, and the expressions of IL-8 and CXCR-I in esophageal tissues were detected by IHC and Radio-immunity. Results The pathological score in model group was significantly higher than the other three groups （ P 〈0.05 ）. Compared with normal group, the expression and content of IL-8 in model group were higher significantly （P 〈0.05, P 〈0.01 ）. Compared with model group, the expression and content of IL-8 in Chinese medicine group and western medicine group were lower significantly （P 〈 0.01 ）. No significant difference was found in expression of CXCR-I among the groups （P 〉 0.05）. Conclusion ＂Dingxiang Jiangqi Decoction＂ can improve the pathological condition in rats with reflux esophagitis. Its mechanism may be include down-regulating the content of IL-8 and relieving the symptoms of esophagitis.
