摘 要:目的制备大鼠视网膜挫伤模型,探讨谷氨酰胺对眼挫伤大鼠视网膜HSP27和NF-κB表达的调节作用。方法SD雄性大鼠45只,5只用于正常对照组,余下40只仿Allen's重击法制作视网膜挫伤模型。将模型制备成功大鼠随机分为两组:模型组和Gln治疗组。每组又分12h,24h,3d,7d四个时间点,每个时间点大鼠5只。各组于给药给水停止后第2天取材,制备石蜡切片。应用免疫组织化学方法检测视网膜组织中HSP27和NF-κB蛋白的表达变化。结果正常组大鼠视网膜内均有HSP27和NF-κB蛋白表达;眼挫伤后HSP27蛋白表达逐渐下降,至3d时表达降至低谷。除12h外,模型组HSP27蛋白表达均比正常对照组低;而NF-κB蛋白表达在眼挫伤后逐渐上升,3d达峰值。除12h外,其余各组NF-κB蛋白表达均比正常对照组高;在Gln组,HSP27蛋白表达均比模型组高,而NF-κB蛋白表达均比模型组低。结论 HSP27和NF-κB参与了眼挫伤视网膜病变过程;谷氨酰胺上调了挫伤视网膜HSP27蛋白表达,下调了NF-κB蛋白表达,这可能是谷氨酰胺对眼挫伤视网膜病变产生保护作用的机理之一。[ Objective ] To establish a retinal contusion model and explore the regulative effect of Glutamine on expression of HSP27 and NF- K B on retina following eye contusion in rats. [ Methods ] Of 45 male SD rats, 5 rats were obtained at random for normal control group.The retinal contusion model was made by modified Allen' s failing strike for the rest 40 rats. The successful model rats were randomLy divided into model group and Gln treat group. Each group is subdivided into 4 groups according to the different timepoint (12h, 24h, 3d, 7d), 5 rats per group. The Gln group animals were injected with Gln intraperitoneally once a day. The model group animals were injected with physiological saline. The left eyes were gotten after stopping giving Gln and physiological saline. Paraffin retinas sections were made and the expressions of HSP27 and NF-κB protein were detected with immunohistochemistry staining. [ Results ] HSP27 and NF-κB protein were expressed in normal groups. The expression of HSP27 protein decreased gradually, and reached minimum at 3 day after ocular trauma in model groups. Except for 12h, the expression of HSP27 protein was lower in model groups than that in normal groups at each timepoint; While the expression of NF-κB protein gradually increased and reached peak at 3 day after ocular trauma in model groups. Except for 12h, the expression of NF-κB protein was higher in model groups than that in normal groups. The expression of HSP27 protein was obviously higher in Gln groups than that in model groups, While the expression of NF- K B protein was obviously higher in Gln groups than that in model groups at 24h, 3day and 7day. [ Conclusion ] HSP27 and NF-κB take part in pathologic process of ocular trauma; Glutamine upregulates the expression of HSP27 and dowrrregulates the expression of NF-κB on retina following ocular trauma, which maybe is the one of protective mechanism that Glutamine performs its protective effcet on ocular trauma.
