摘  要:目的:通过对小半夏加茯苓汤高效液相色谱(HPLC)条件的考察,建立复方止呕效用成分,即鸟苷、盐酸麻黄碱、芦丁、6-姜酚、8-姜酚、10-姜酚、去氢茯苓酸及茯苓酸8种成分的特征图谱方法学。方法:分别考察复方50%醇提液HPLC分离的流动相、色谱柱、洗脱梯度及检测波长四方面因素。以RP-HPLC法梯度洗脱,Phenomenex Gemini C18色谱柱(5μ,250mm×4.6mm);乙腈-0.1%磷酸水溶液洗脱流动相;检测波长236nm;柱温40℃;流速1mL/min。结果:在一定范围内,鸟苷、盐酸麻黄碱、芦丁、6-姜酚、8-姜酚、10-姜酚、去氢茯苓酸及茯苓酸峰面积积分值Y与进样量X线性关系良好,加样回收率分别为98.33%、97.26%、97.81%、96.60%、98.01%、97.09%、96.46%和95.90%。结论:该方法操作简便、准确、重复性好,为复方质量标准评价提供了依据。Objective:Through the investigation of Xiaobanxia plus Fuling Decoction by HPLC,constitute the compound fingerprint profiles of chromatogram of Guanosine Ephedrine,Hydrochloride,Ruti,6-Gingerol,8-Gingerol,10-Gingerol,Dehydro-pachymic acid and Pachymic acid.Methods:Compound fingerprint profiles of chromatogram methodology was established by individually investigating four factors,containing compound 50% alcohol extract of HPLC separation,the mobile phase chromatographic column,elution gradient and detection wavelength.The method was performed on Phenomenex C18 column,using acetonitrile-0.1% phosphoric acid solution,gradient elution,on the condition of 220nm detection wavelength as gradient eluents.Results:To some extent,for Guanosine,Ephedrine,Hydrochloride,Rutin,6-Gingerol,8-Gingerol,10-Gingerol,Dehydro-pachymic acid and Pachymic acid,a good linearity relationship was exhibited between the peak areas Y and injection volume X.The average recovery was 98.33%,97.26%,97.81%,96.60%,98.01%,97.09%,96.46% and 95.90%.Conclusion:The method is simple and accurate,with a good reproducibility.It can be used as an analysis method for compound quality standard.
