“寒淫”致病对大鼠基因表达及相关通路影响的实验研究

摘  要:寒淫致病以及中药干预下,寻找大鼠差异基因表达及相关信号通路,探讨寒淫对大鼠免疫功能的影响。方法:分析正常组与寒淫组、正常组与麻黄细辛附子汤药组比较筛选差异表达基因,采用了基因功能、信号通路分析等方法探讨寒淫致病对大鼠免疫功能的影响,并作RT-PCR验证。结果:通过差异表达基因作高级筛选,得到共同的差异表达基因21条及通路10条,其中与寒淫致病密切相关的免疫功能基因及其通路有RT1-Ba基因与抗原处理和递呈通路。结论:结合差异表达基因谱的分析,提示寒淫致病与细胞因子等有密切的相关性,为下一步的研究寒淫致病提供了重要的指示性研究方向。Objective:To establish a cold pathogen-induced disease in animal models,and by the intervention of traditional Chinese medicine to find differences in gene expression in rats and related signaling pathways of the cold excessive immune functions.Methods:The different gene expressions in the normal group,cold group,and drug group were analyzed by gene function,signaling pathway analysis methods and RT-PCR.Results:By screening differentially expressed genes for high-level,there were 21 differentially expressed genes and 10 pathways,which was closely related with the cold excessive immune function of disease genes and RT1-Ba gene and antigen processing and the delivery was included.Conclusion:The combination of differentially expressed genes in comparative analysis of spectra suggests that excessive cold pathogen and immune function are closely related.The result may provid an important way to the further study of pathogenic cold research in Chinese medicine.
