摘 要:目的:探讨固肠方对化疗药物致肠黏膜屏障损害的保护作用及可能机制。方法:将雄性BALB/c品系小鼠随机分为正常组、模型组、西药对照组、低剂量固肠方组、中剂量固肠方组、高剂量固肠方组。采用化疗药物氟尿嘧啶致肠黏膜屏障损害法造模,并予药物干预治疗2周。分别于治疗前后检测各组小鼠体质量、回肠黏膜厚度,及血清中肿瘤坏死因子-α(TNF-α)、白介素-6(IL-6)表达水平;并在光镜下观察回肠、结肠组织结构病理变化。结果:中剂量固肠方组能明显改善小鼠体质量减轻状况(P<0.05);中剂量、高剂量固肠方组回肠、结肠病理变化较模型组明显改善,其中高剂量组肠绒毛高度明显高于模型组(P<0.05),且肠绒毛高度变化、肠黏膜病理改善情况与中药给药浓度呈正相关;各药物组对TNF-α、IL-6表达水平均有降低趋势,其中中剂量组有显著性差异(P<0.05)。结论:中、高剂量固肠方对化疗药物氟尿嘧啶致肠黏膜屏障有显著的保护作用,其机制可能与下调TNF-α、IL-6的表达,减少化疗药物致肠道的炎症反应,从而减轻肠黏膜损伤有关。Objective: To discuss the protective effect and mechanism of "Guchang Prescription" on impaired intestinal mucosal bahlet caused by chemotherapy. Methods: Male BALB/c mice were randomized into normal group, model group, western medicine control group and " Guchang Prescription" groups ( small, medimn and large dosages). The models were established by chemotherapy with 5-Fluorouracil. Each group was intervened by different drugs for 2 weeks. The weight of mice, thickness of intestinal umcosa, expression of TNF-α and IL-6 were detected before and after treatment ; the pathological changes of ileum and colon were observed by light microscope. Results: The weight lose in medium dose " Guchang Prescription" group was improved ( P 〈 0.05 ) ; the pathological changes of ileum and colon in medium dose and large dose groups were improved, and the height of intestinal villus was higher in large dose group than that in model group ( P 〈 0.05 ) ; the height of intestinal villus and improvement of intestinal mucosa were associated with concentration of Chinese medicine; expression of TNF-α and IL-6 in medicine groups were decreased, with significant difference in medium " Guchang Prescription" group ( P 〈 0. 05 ). Conclusion : Medium dose and large dose of " Guchang Prescription" has significant protective effect on impaired intestinal mueosal barrier caused by chemotherapy, and the mechanism may be assoeialed with regulation the expression of TNF- c~ and IL-6, and reduction of inflammatory reaction so as to alleviate the impaired intestinal mueosa.
