摘　　要：目的建立测定西罗莫司滴眼液含量的高效液相色谱方法。方法采用Diamonsil C18(250 mm×4.6 mm,5μm)为色谱柱;乙腈-甲醇-水(7.5∶62.5∶30,v/v/v)为流动相,柱温50℃,检测波长276 nm,流速1.2 ml/min。结果在20～80μg/ml浓度范围内,西罗莫司峰面积和浓度呈良好的线性关系(r=0.999 7),平均回收率为100.1%,RSD为0.3%,精密度高,耐用性好,样品溶液可以在室温环境下24 h内稳定。结论本方法专属性强,操作简便,结果准确,可用于测定西罗莫司滴眼液的含量。Objective To establish an HPLC method for the determination of sirolimus eye drops. Methods The separation was performed on Diamonsil C18 （250 mm × 4.6 mm, 5 um）column, the mobile phase was acetonitrilemethanol-water （7.5 ： 62.5 ： 30） with the flow rate of 1.2 ml/min, the detection wavelength was at 276 nm and column temperature was 50 ℃. Results The calibration curve of sirolimus was linear within the concentration range of 20 -80 ug/ml, r =0. 999 7. The average recovery rate of sirolimus was 100. 1% （RSD = 0.3% ）. Conclusion The method is simple, quick and specific. It can be used to determine the content of sirolimus eye drops.
