摘 要:目的首次考查何首乌提取物抗抑郁活性。方法采用昆明KM雄性小鼠,每组10只。何首乌水提物(HW)、何首乌75%乙醇提取物(HE)及阳性药盐酸氟西汀(FH)均以0.5%羧甲基纤维素纳(CMC-Na)溶解或混悬至所需给药浓度,HW和HE剂量均分别为1.5、3 g/kg,FH剂量为10mg/kg,模型组给予同等体积的0.5%CMC-Na,各组均连续灌胃给药7 d,每天1次,末次给药1 h后,开展小鼠悬尾实验(TST)、强迫游泳实验(FST)和开场实验(OFT)测试。结果 HW(1.5、3 g/kg)可使小鼠在TST及FST的不动时间均显著性缩短(P<0.05),呈现出良好的量效关系,且与阳性药FH相比无显著性差异(P>0.05),而HE(1.5、3 g/kg)对小鼠在TST及FST的不动时间均无显著性差异(P>0.05);各组小鼠OFT穿格次数比较,均无显著性差异(P>0.05)。结论首次证实何首乌水提物具有抗抑郁活性,而其醇提物在既定给药方案下无抗抑郁活性。Objective To examine for the first time the antidepression activity of extract of Heshouwu (Radix Polygoni Multiflori). Methods Male Kunming mice were divided into groups (each n = 10). The aqueous extract and ethanol (75 % ) extract of Heshouwu and fluoxetine hydrochloride (FH) were all dissolved or suspended in 0.5% CMC-Na to reach to desired concentration. The doses of aqueous extract were 1.5 g/kg and 3 g/kg respectively, and ethanol extract, also 1.5 g/kg and 3 g/kg respectively, and dose of FH was 10 mg/kg. Equivalent dose of 0.5% CMC-Na was given to model group. All groups were intragastrically given corresponding medicinals once a day for 7 days. After the last time of giving medicinals for one hour, the tail suspension test( TST), forced swimming test(FST) and open field test(OFT) were conducted in mice. Results The aqueous extract ( 1.5 g/kg and 3 g/kg) shortened significantly the dead time in TST and FST ( P 〈 0.05 ) showing a good dose-effect relation. There was no significant difference compared with FH(P 〉 0.05 ). The ethanol extract( 1.5 g/kg and 3 g/kg) had no significant differences in the changes of the dead time in TST and FST( P 〉 0.05 ). The comparison of crossing times in OFF was not different among all groups ( P 〉 0.05 ). Conclusion It is proved for the first time that the aqueous extract of Heshouwu has antidepression activity, while its ethanol extract had no under an established medicinal administration plan.
