摘  要::目的 观察内含子对脂质体介导的转基因表达的影响。方法 首先构建含内含子intronA、IL 2基因、IFN γ基因的质粒表达载体 ,然后经阳离子脂质体LipofectAMINE介导转染小鼠成纤维细胞NIH3T3。生物活性法检测IL 2基因或IFN γ基因表达水平。结果 含内含子的载体其IL 2或IFN γ表达水平高于不含内含子的载体(P <0 0 5 )。Objective: To observe the effect of intron on gene expression after liposome mediated transfection. Methods: First, the plasmid expression vectors carrying intron A, IL 2 gene , IFN γ gene, p53 were constructed. And then, these genes were transfected into NIH3T3 cells with LipofectAMINE. The expression level of IL 2 and IFN γ in culture supernatant was examined by means of biological activity. Results: Higher gene expression level of IL 2 or IFN γ in intron carrying vectors was observed as compared with non intron vectors. Conclusion: The gene expression level may be enhanced by intron A after liposome mediated transfection.
