摘  要:目的 :研究ACL和MCL细胞在mRNA表达水平上的差别。方法 :培养人ACL和MCL细胞 ,提取总RNA ,用mRNA差异显示技术寻找ACL和MCL细胞在mRNA表达水平上的差异表达带 ,将有差异的条带回收、扩增、克隆化及ReverseNorthern杂交 ,筛选出阳性克隆进行cDNA测序并进行NCBI基因数据库分析。结果 :在ACL和MCL细胞之间找到 6个差异表达的基因 ,分别为C4M2 ,C1A1,A4M1,A1A2 ,G4A1,C7A2。结论 :ACL和MCL细胞在基因表达水平上有区别 ,已找到的基因可作为探针筛选cDNA文库进一步研究其功能。Objective To study the differences between ACL and MCL cells at the level of mRNA expression. Methods Human ACL and MCL cells were cultured, and total RNA was extracted. mRNA differential display was used to compare the differences between the two groups. The differentially displayed gene bands were isolated, reamplified and cloned for Reverse Northern Hybridization. The positive cloned genes were sequenced and compared to GenBank database analysis using Basic Local Aligment Search Tool of NCBI. Results Six differentially displayed genes were found. These genes were named C4M2, C1A1, A4M1, A1A2, G4A1, C7A2. Conclusion There were some differences between ACL and MCL at the level of mRNA gene expression. These cloned genes can be used as probes to screen the cDNA library and study the gene functions.
