摘 要:目的观察原发性干燥综合征(pSS)患者外周血单核源性树突状细胞在聚肌苷酸胞苷酸(polyI∶C)刺激下释放干扰素等炎性因子的能力。方法用白细胞介素(IL)-4和粒细胞-巨噬细胞集落刺激因子从pSS患者外周血单核细胞中诱导出树突状细胞,并用50μg/mL polyI∶C进行刺激,在刺激后第6、12、24小时以ELISA法检测细胞培养基上清液中干扰素(IFN)-α、IFN-β、肿瘤坏死因子-α(TNF-α)、IL-1β、IL-6、IL-8、IL-12和IL-23的浓度,以RT-PCR检测树突状细胞内Toll样受体3(TLR3)分子的表达。结果在polyI∶C作用下,pSS患者单核源性树突状细胞释放IFN-α、IFN-β及TNF-α等因子的能力显著强于来自健康个体的单核源性树突状细胞。虽然pSS患者及健康个体单核源性树突状细胞内TLR3的表达在polyI∶C刺激作用下均随时间逐渐降低,但pSS患者TLR3的降低相对缓慢。结论 pSS患者单核源性树突状细胞对polyI∶C刺激呈现高反应性,且这种高反应性可能与TLR3信号作用异常有关。单核源性树突状细胞的这种特征可能与pSS的发病机制有关。Objective To investigate the potential correlation between pathogenesis of primary Sjrgren's syndrome (pSS) and the funetion of monocyte -derived dendritic cells. Methods Monocytes from 11 pSS patients and 11 healthy controls were isolated and induced to dendritic cells in the presence of IL - 4 and GM - CSF. Polyinosinic polycytidylic acid (polyI: C, 50 μg/mL) was used to simulate the monoeyte -derived dendritic cells. Serial cytokines and TLR3 mRNA were assessed by ELISA and RT -PCR, respectively, at the 6th, 12th and 14th hour after polyI: C treatment. Results Significantly more IFN - α, IFN -β and TNF - α were produced in monocyte - derived dentritic cells from pSS patients when challenged with polyI: C. Although TLR3 mRNA expression was down -regulated by polyI: C stimulation, it was impaired in pSS patients. Conclusion The Hypersensitivity to polyI: C, which may associated with TLR3 abnormality, of the monocyte - derived dendritic cells from pSS patients was identified. The abnormality of monoeyte - derived dendritic cells from pSS patients may involved in the pathogenesis of pSS.
