摘　　要：目的白细胞介素17在多种心血管疾病中如自身免疫性心肌炎、病毒性心肌炎、扩张型心肌病、不稳定型冠心病等表达增加,在心血管病慢性炎症性过程起到了一定作用。分析白细胞介素17在慢性心力衰竭大鼠心肌中的表达特点。方法采用腹主动脉缩窄术构建大鼠慢性心力衰竭模型。将62只雄性SD慢性心力衰竭大鼠模型随机分14d组、28d组、56d组及112d组,另设假手术组大鼠10只。采用酶联免疫吸附测定法(ELISA)、免疫荧光激光共聚焦技术(LSCM)及蛋白免疫印迹法(Western blot)检测各组大鼠心肌IL-17的表达。结果造模成功率为58.06%。ELISA、Western blot、激光共聚焦的结果均表明在大鼠慢性心力衰竭的发生发展过程中,IL-17在心肌组织大量表达,且随着时间变化而逐渐升高,在112d表达最为显著。结论 IL-17作为一种强力的促炎症因子在慢性心力衰竭大鼠心肌组织中大量表达,可能与慢性心力衰竭后的心室重塑及心律失常的发生有关。Objective Expression of iterleukin-17 in many cardiovascular disease increases such as autoimmune myocarditis,viral myocarditis,dilated cardiomyopathy and unstable coronary artery disease.Iterleukin-17 plays an important part in chronic inflammatory process of cardiovascular disease.To study the expression of interleukin-17 of myocardium in rats with chronic heart failure.Methods Abdominal aorta was constricted to induce chronic heart failure in rats.82 rats were randomly divided into six groups according to different observation times：14,28,56,112 days groups.And 10 rats were selected as control.The expression of interleukin-17 was examined by enzyme linked immunosorbent assay,laser confocal scanning microscope and Western blot.Results Modeling success rate was 58.06%.With the development of myocardial infarction,interleukin-17 was abundantly expressed in myocardial and gradually increased over time,and interleukin-17 in the 112 days group was expressed the most significant.Conclusions Interleukin-17 as a potent pro-inflammatory factor can strongly express in rat myocardium with chronic heart failure,which may be related to the incidence of ventricular remodeling and arrhythmia.
