青海省流感病毒病原学监测及H3亚型血凝素基因变异研究

摘  要:目的监测青海省2007~2008年度季节性流感病毒型与亚型分布,了解2007~2008年度部分A/H3N2分离株血凝素(HA)基因变异情况。方法采集监测哨点医院流感样病例的鼻咽双拭子标本,接种MDCK细胞,采用标准抗血清鉴定阳性分离株型与亚型,并对2007~2008年度部分H3亚型流感病毒进行HA基因测序,分析HA基因变异情况。结果 2007~2008年度在哨点医院送检标本中共分离到144株流感病毒,其中124株为A/H3N2,B型20株,型与亚型有明显分布强弱特征。H3亚型HA序列分析,青海省6株分离株与2008、2009年部分省份分离株及江西同期分离株接近,与WHO2008~2009年度疫苗推荐株相近。结论 WHO推荐的2006~2007年度疫苗株A/Hiroshima/52/2005滞后于在我省流行的流感病毒毒株。OBJECTIVE To understand the type and subtype distribution of influenza virus and hemagglutinin (HA) gene variation of strained H3N2 from 2007 to 2008 in Qinghai Province. METHODS The nasal swabs specimens of sentinel hospital ILI from 2007 to 2008 were collected and inoculated with MDCK cells, and then identificated by hemagglutination inhibition (HI) method. The HA genes of six strains subtype H3N2 virus were sequenced and the HA gene variation was analyzed. RE- SULTS It Was shown that 144 stains influenza virus were isolated, including 124 stains subtype H3N2 and 20 stains type B. The distribution of type and subtype is significantly different. After the HA gene sequence analysis of subtype H3N2, we knew that the sequences of six strains isolated form Qinghai Province from 2007 to 2008 were similar to that of stains isolated from Jiangxi Province and some other provinces from 2008 to 2009, and similar to vaccine strains recommended by WHO from 2008 to 2009. CONCLUSION The vaccine strains, A/Hiroshima/52/2005, recommended by WHO from 2006 to 2007 drop behind the pandemic influenza virus stains in Qinghai Province.
