摘 要:目的观察可诱导共刺激分子(ICOS)在BXSB狼疮小鼠肾脏中的表达,探讨ICOS在狼疮肾炎发病机制中的潜在作用。方法选取8周龄及16周龄雄性BXSB小鼠各6只,并以8周龄C57BL/6雄性小鼠6只为正常对照,用免疫组织化学及实时荧光定量PCR方法,检测ICOS在小鼠肾脏中的表达水平。结果正常对照组C57BL/6小鼠肾组织ICOS染色少许肾小管细胞呈浅棕色;8周龄及16周龄BXSB小鼠肾脏组织ICOS强阳性表达,细胞呈深棕色着色,分布于肾小管。8、16周龄BXSB组小鼠肾组织内ICOS mRNA表达水平[(6.43±0.92),(9.48±1.30)]均较正常对照组(4.58±0.63)增加,差异有统计学意义(均P<0.01);16周龄BXSB组小鼠的ICOS mRNA表达水平较8周龄BXSB组增加,差异有统计学意义(P<0.05)。结论肾组织ICOS的分布及表达增加可能是BXSB小鼠狼疮肾炎发病的重要机制之一。Objective By analysis of the expression of inducible costimulator (ICOS) in the kidney of BXSB mice, this study was trying Io clarify the potential role of ]COS in the pathogenesis of lupus nephritis in BXSB mice. Methods ICOS was detected in control mice, 8-week and 16-week BXSB male mice by u- sing immunohistochemistry and real-time PCR. Results ICOS immunostaining in control mice was light brown and distributed in few renal tubules; but in & and 16-week BXSB mice, the ICOS immunostaining was much stronger as dark brown than that in control mice. The mRNA expression of ICOS was signifi cantly increased in renal tissues from 8- and 16-week BXSB mice as compared with that in the control mice (6. 434-0.92 and 9. 484-1.80 vs 4.58!0.63, P^0.01), and furthermore, there was significantly differ- ence in ICOS mRNA expression between 8- and 16-week BXSB mice (P〈0.05). Conclusion Distribution and up regulation of ICOS may constitute a phritis in BXSB mice.
