摘 要:目的观察L-选择素(L-sellectin,SELL)基因rs4987318单核苷酸多态性(single nucleotide polymor-phism,SNP)与2型糖尿病(type 2 diabetes mellitus,T2DM)的相关性和生化指标的变化。方法应用荧光标记单碱基延伸及寡核苷酸微阵列芯片杂交技术检测SELL基因rs4987318多态性;采用全自动生化分析仪检测血脂及C-反应蛋白(C-reactive protein,CRP)水平。结果 T2DM组CC基因型频率明显高于健康对照组(P<0.05)。T2DM组的甘油三酯(triglyceride,TG)、载脂蛋白B(apolipoprotein B,Apo B)和CRP水平显著高于对照组(P<0.05),高密度脂蛋白胆固醇(high-density lipoprotein cholesterol,HDL-C)和载脂蛋白A1(apolipoprotein A1,Apo A1)水平显著低于对照组(P<0.05)。T2DM组TG的AA、CA、CC型,Apo B的CC型和CRP的CA、CC型的水平显著高于对照组的相同基因型(P<0.05);HDL-C的AA、CA、CC型和Apo A1的AA、CA型水平显著低于对照组的相同基因型(P<0.05)。结论 SELL基因rs4987318多态性可能与T2DM的易患性、患者血脂代谢紊乱和血管炎症的发生有关。Objective To investigate the association between rs4987318 single nucleotide polymorphism(SNP) in L-sellectin(SELL) gene and type 2 diabetes mellitus(T2DM) and the clinical biochemical indexes.Methods The rs4987318 mutations in the SELL gene were analyzed with allele-specific fluorogenic oligonucleotide probes in an assay combining extension and hybridization.The levels of lipid and C-reactive protein(CRP) were detected by automatic biochemical modular system.Results The frequencies of CC genotype of T2DM group was significantly higher than that of the control group(P〈0.05).In T2DM group,the serum levels of triglyceride(TG),apolipoprotein B(Apo B) and CRP were higher,and the high-density lipoprotein cholesterol(HDL-C) and apolipoprotein A1(Apo A1) were lower than that in the control group(P〈0.05).TG levels of AA,CA and CC genotype,Apo B level of CC genotype,CRP levels of CA and CC genotypes were significantly higher in T2DM group than that of the same genotypes in the control group(P〈0.05).HDL-C levels of AA,CA and CC genotypes,Apo A1 levels of AA and CA were significantly lower in T2DM group than that of the same genotypes in the control group(P〈0.05).Conclusion Rs4987318 polymorphism in SELL may be associated with T2DM susceptibility,lipid metabolism disorder and vessel inflammation of T2DM patients.
