中药体外防治甲型H1N1流感病毒实验研究

摘　　要：目的研究中药防治流感的体外效果。方法采用鸡红细胞凝集实验以及MTT法观察三种常见的代表性预防流感的方药("扶正"预防类-玉屏风制剂;祛邪预防类-银翘散制剂;外用芳香辟秽类制剂-冰香散)对流感病毒A/FM/1/47(H1N1)鼠肺适应株的预防作用。结果冰香散在最小无毒浓度15.6μg/ml时对流感病毒A/FM/1/47(H1N1)鼠肺适应株有良好的预防作用。银翘散和玉屏风散仅在毒性浓度下对流感有预防作用,无毒浓度下均不能抑制病毒。阳性对照药利巴韦林在0.25～2 mg/ml浓度内均有较好的抑制流感病毒作用。冰香散提前加药,冰香散对病毒的半数抑制浓度(IC50)为5.89μg/ml,治疗指数(TI)为3.28;冰香散与病毒同时加药,冰香散挥发油对流感病毒的直接抑制作用的IC50为9.56μg/ml,TI为2.02。而先加流感病毒吸附后再加冰香散,则未能对流感病毒有抑制作用,其治疗指数TI小于0.05。结论冰香散体外对甲型流感病毒FM1有预防作用。玉屏风散制剂和银翘散制剂仅在毒性浓度下对流感有预防作用,无毒浓度下均不能抑制病毒。其防治流感病毒的作用可能通过体内免疫机制而实现。Objective To evaluate the preventive effects of three Chinese medicines against influenza A （H1N1） in vitro. Methods The mouse lung- adapted influenza virus strain A/FM/1/47（H1N1 ） was cultured in madin darby canine kidney （MDCK） cells and chicken erythrocyte agglutination test and MTTassay were performed to evaluate the influenza- preventive effects of three Chinese medicines, including Jade Screen Powder Preparation with functions of strengthening body resistance, Yinqiao Powder Preparation with functions of eliminating pathogens and Bing- xiang Powder Preparation with functions of dis- pelling turbidness by aromatic, essential oils. Results Bing xiang Powder Preparation exhibited good preventive effects against the mouse lung- adapted influenza virus A/FM/1/47（H1N1 ） at the minimum non - toxic concentration of lfi. 6/~g/ml, while Yinqiao Powder Preparation and Jade Screen Powder Preparation produced inhibitory effects on the virus only at toxic con- centrations. The positive control drug, Ribavirin, could significantly inhibit influenza virus at concentrations between 0.25 μg/ML and 2 μg/ml. When Bing- xiang Powder Preparation was administered to the culture before the influenza virus exposure, the median inhibitory concentration （ICs0） was ft. 89μg/ml and the therapeutic index （TI） was 3.28. When administrated upon vir- us exposure, it produced a direct inhibitory effect on the virus, with an ICs0 of 9.56μg/ml and a TI of 2.02. However, when it was administered after the influenza virus was adsorbed, it failed to inhibit the virus, with a TI less than 0.05. Conclusions Bing- xiang Powder Preparation can produce preventive effects against influenza virus in vitro. Jade Screen Powder Preparation and Yinqiao Powder Preparation can inhibit influenza virus in vitro only at toxic concentrations. They might prevent against in- fluenza virus by immune mechanisms in vivo.
