摘 要:目的:观察回药爱康方对小鼠肿瘤生长的影响及对突变型p53蛋白(p53mt)表达的影响。方法:将55只C57小鼠常规接种Lewis肺癌细胞,24h后随机分为荷瘤模型组、化疗组、回药低剂量组、回药低剂量加化疗组、回药高剂量加化疗组。治疗后观察各组小鼠的脾脏、胸腺、瘤体质量,免疫组化法检测各组p53mt的表达差异。结果:①除化疗组外,其余各组小鼠体质量与荷瘤模型组比较差异均有统计学意义(P<0.05);②回药高剂量加化疗组抑瘤率为71.00%,与荷瘤模型组比较,各组瘤质量差异有统计学意义(P<0.05);③回药高剂量加化疗组p53mt表达阳性率为36.36%,与模型组比较有统计学意义(P<0.01);④与化疗组比较,各组小鼠脾脏指数及胸腺指数差异均有统计学意义(P<0.01)。结论:回药协同化疗抑瘤效果及下调突变型p53的表达效果优于单纯化疗,且回药爱康方可以降低化疗后的免疫抑制,这可能是回族医药治疗肿瘤的新靶点。Objective:To observe the Hui medicine Aikang Fang's(HAF) effects on tumor growth in mice and changes of mutant p53(mtp53) expression.Method:55 normal C57 mice were inoculated with Lewis lung cancer cells.After 24h,the mice were randomly divided into model group,chemotherapy group,the low-dose group,the low-dose with chemotherapy and high-dose with chemotherapy group.After treatment,the spleen,thymus and tumor mass of all rats were tested.Expressions of mtp53 in all groups had a significantly different using immunohistochemical method.Result:① In addition to chemotherapy group,the difference between the rest groups and model group was significant in body weight(P〈0.05);②Inhibition rate of high-dose group was 71%.Compared with tumor-bearing model group,the difference of tumor weight in all groups were significant(P〈0.05);③Mtp53 expression rate of high-dose with chemotherapy group was 36.36%.Compared with tumor-bearing model group,HAF high-dose with chemotherapy group was significantly different(P〈0.01);④Compared with the chemotherapy group,spleen index and thymus index differences of the rest group mice were statistically significant(P〈0.01).Conclusion:Inhibition of HAF with chemotherapy and effects of reducing the expression of mtp53 were better than chemotherapy alone,and HAF could reduce immune suppression after chemotherapy,it may be a new target of treatment cancer.
