摘  要:目的研究脓毒症休克大鼠下丘脑和延髓中胱硫醚B合酶(CBS)mRNA的表达状况。方法12只雄性SD大鼠随机分为假手术组(n=6)和脓毒症休克组(n=6),脓毒症休克模型采用盲肠结扎穿孔术(CLP)。术后大鼠测血压、观察一般表现,确定脓毒症休克模型是否成功;逆转录PcR(RT-PCR)和实时荧光定量PCR(real-timePCR)检测下丘脑、延髓中CBS基因表达。结果脓毒症休克组大鼠下丘脑及延髓均有CBS的表达。与假手术组比较,脓毒症休克大鼠延髓CBSmRNA表达明显升高(P〈0.05);下丘脑未有明显变化(P〉0.05);与假手术组下丘脑比较,假手术组延髓CBS表达显著降低(P〈0.05);与脓毒症休克组下丘脑比较,脓毒症休克组延髓CBS表达显著升高(P〈0.05)。结论脓毒症休克大鼠脑组织CBSmRNA表达上调,可能通过延髓或下丘脑机制参与脓毒症休克的发病机制。Objective To investigate the expression of cystathionine beta- synthase (CBS) mRNA in brain tissue ( hypothalamus, medulla oblongata) of the septic shock rat. Methods Twelve male SD rats were randomized into sham group and septic shock group, in which the sepsis model adopts cecal legation perforation method (CLP). After surgery, in order to define the septic shock, blood pressure was assayed, the change of behavior was observed. The expression of CBS in brain tissue (hypothalamus, medulla oblongata ) was detected by reverse transcription PCR ( RT - PCR ) and quantitative fluorescence polymerase chain reaction ( real - time PCR). Results The expression of CBS was discovered in hypothalamus and medulla oblongata of septic shock rats. Compared with sham group, it is shown that the expression of CBS in medulla oblongata of CLP rats was significantly increased(P 〈 0.05), but no significantly change was observed in the hypothalamus (P 〉 0. 05 ). Compared with hypothalamus of sham group, it is shown that the expression of CBS in medulla oblongata of CLP rats was significantly decreased(P 〈 0. 05 ). Compared with hypothalamus of CLP group, it is shown that the expression of CBS in medulla oblongata of CLP rats was significantly increased(P 〈 0.05 ). Conclusion The expression of CBS mRNA in brain tissue of septic shock rats are up - regulated, it is likely to get through the mechanism of hypothalamus and medulla oblongata, which probably involved in the pathogenesis of septic shock.
