摘  要:目的探讨血小板聚集诱导剂花生四烯酸(AA)和二磷酸腺苷(ADP)的储存条件。方法随机收集101例门诊体检健康志愿者静脉血标本,枸橼酸钠抗凝,离心分离富含血小板血浆(PRP)和贫血小板血浆(PPP),采用血浆比浊法,分别用不同储存条件AA和ADP作为诱导剂,上机测血小板聚集率。结果 AA-50℃储存0月和7月诱导血小板聚集率分别为(47.9±37.5)%和(47.3±37.2)%,ADP-50℃储存0月和13月诱导血小板聚集率分别为(66.2±15.7)%和(67.9±17.2)%(均P>0.05);-50℃解冻后储存冰箱5℃0、24h,AA诱导血小板聚集率分别为(47.8±38.8)%和(46.1±38.3)%,ADP诱导血小板聚集率分别为(67.1±25.8)%和(68.8±25.9)%(均P>0.05)。结论 -50℃储存AA 7月、ADP13月、AA和ADP-50℃解冻后储存冰箱5℃24h,对血小板聚集无影响。Objective To explore the storage condition of arachidonic acid (AA) and adenosine diphosphate (ADP) for platelet aggregation. Method Venous blood sample of outpatient volunteers were collected (n = 101 ) randomly and anticoagulaged sodium citrate, and platelet-rich plasma(PRP) and platelet-poor plasma was obtained by centrifuging the blood, then platelet aggregation rate of the plasma were detected with inducer of AA and ADP under the different storing conditions by plasma turbidimetry. Result When the inducer of AA and ADP were stored for 0 and 7 months, 0 and 13 months in a -50℃ refrigeration respectively, the platelet aggregation rates were (47.9±37.5 ) % and (47.3 ± 37.2) %, ( 66.2 ±15.7 ) % and (67.9± 17.2) % respectively (P 〉 0.05 ). When AA and ADP after thawing stored for 0 h and 24 h at 5 ℃, the rates were (47.8±38.8)% and (46.1±38.3)%, (67.1±25.8)% and (68.8±25.9)% respectively(P 〉0.05). Conclusion The inducers of AA and ADP store for 7 months and 13 months in a -50℃ refrigeration and AA and ADP after thawing store for 24 h at 5℃ respectively, the effect on the testing result of platelet aggregation is not significant.
