摘  要:目的研究夏枯草总三萜(TTP)对大鼠急性肝损伤的保护作用。方法 60只SD大鼠随机均分为对照组、模型组、TTP(62.5、125、250 mg/kg)3个剂量组和联苯双酯(200mg/kg)组。各给药组及联苯双酯组预防给药6 d,第6天给药结束后腹腔注射50%CCl4(2 ml/kg)诱导大鼠急性肝损伤模型,观察TTP对大鼠血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)水平的影响以及对肝匀浆中丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)水平的影响;肝组织HE染色;RT-PCR法、West-ern blot法检测肝组织细胞色素P4502E1(CYP2E1)表达。结果 TTP各剂量组可降低急性肝损伤大鼠血清中ALT、AST活性,降低肝匀浆MDA的水平,升高SOD、GSH-Px的水平,抑制肝组织中CYP2E1的表达;同时,病理组织学检查结果显示TTP各剂量组大鼠肝组织病变程度明显减轻。结论TTP对CCl4致大鼠急性肝损伤具有一定的保护作用,其机制可能与抗机体脂质过氧化、抑制CYP2E1表达有关。Objective To investigate the hepatoprotective effect of total triterpenoids of Prunella vulgaris L. (TrP) on rats. Methods Sixty rats were randomly divided into six groups as follows: control group, CCl4 group, TTP (62.5, 125 and 250 mg/kg) groups and bifendate (200 mg/kg) group. Rat acute liver injury model was induced by peritoneal injection of 50% CCl4 (2 ml/kg, dissolved in corn oil). Rats in therapeutic groups were pretreated with TTP or bifendate six days before CCl4 was given. The effects of TTP on serum levels of alanine aminotransferase (ALT) and aspartate aminotransferase ( AST), hepatic malondialdehyde (MDA) content, glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) activities in different groups were investigated. In addition, his- topathologic changes of the liver were detected with hematoxylin-eosin (HE). CYP2E1 protein and mRNA expressions in liver tissue were evaluated by Western blotting, immunohistochemical staining and transcriptase-polymerase chain reaction, respectively. Results TTP effectively decreased serum ALT, AST levels, hepatic MDA content, and restored hepatic SOD and GSH-Px activities in acute liver injury rats. Furthermore, TFP reduced CYP2E1 expression at both protein and mRNA levels in a dose-dependent manner. Conclusion TTP had protective and therapeutic effect on rat liver injury, which might be associated with its antioxidant properties and inhibition of CYP2E1 activation.
