摘 要:目的探讨双肾一夹大鼠肥厚心肌组织中Calsarcin-1的变化及替米沙坦对其的影响。方法清洁级♂SD大鼠40只,随机分为假手术组(n=10)、两肾一夹(2K1C)组(n=15)和替米沙坦组(n=15)。2K1C组及替米沙坦组用两肾一夹(two kidney one clip,2K1C)法制备高血压性心肌肥厚模型,替米沙坦组以替米沙坦(10 mg.kg.d-1)灌胃干预,2K1C组采用等量蒸馏水灌胃。在术后第4、8周末,超声心动图观察心脏结构及功能,腹主动脉插管测血压,处死大鼠后取心脏,用Western blot方法检测不同时期心肌组织Cal-sarcin-1蛋白表达水平。结果①与假手术组相比,2K1C组大鼠血压于术后4周明显升高,心肌肥厚明显(P<0.01);替米沙坦组血压较2K1C组明显降低,心肌肥厚亦明显减轻(P<0.01);②与假手术组相比,2K1C术后第4及第8周心肌Calsarcin-1表达水平明显下降(P<0.01),替米沙坦灌胃4周后,心肌中Calsarcin-1表达水平较未灌胃组升高(P<0.05),灌胃8周后,心肌中Calsarcin-1表达水平较4周前稍有上升,但二者之间差异无显著性(P>0.05)。结论Calsarcin-1表达下调参与了高血压性心肌肥厚的发生发展;替米沙坦有效降压、并上调Calsarcin-1表达水平,这可能是其逆转心肌肥厚的机制之一。Aim To test Calsarcin-1 level in different stages of cardiac hypertrophy by renovascular hypertensive (2KI C, two-kidneys, oneclip) rat models, and to explore the mechanism of cardiac hypertrophy and the effect of telmisartan. Methods Forty male SpragueDawly rats were randomly divided into sham operation group (n =10), 2 kidney 1 clip (2K1C) group (n = 15) and telmisartan group ( n = 15). 2KIC hyperten- sive rats were established by chronic partial occlusion of left renal artery. Before and after four and eight weeks of the operation, blood pressure was tested by arterial catheterization, the myocardial structure and function was obtained by ultrasonography, and Calsarcin-1 expression level in myocardium was compared by Western blot. Results (1) Eight weeks later, the blood pressure and the thickness of left ventricle in 2K1C group were significantly higher than those of sham group (P 〈0. 01 ). Compared with 2K1C group, the blood pressure and the the thickness of left ventricle were significantly decreased in telmisartan group (P 〈0. 01). (2) Compared with sham group, the Cal- sarcin-1 level was markedly increased in 2K1C group (P 〈 0.01 ) , and after being treated for 4 weeks, the Calsarcin-1 level was higher than that of untreated group (P 〈 0. 05), but Calsarein-1 level was not markedly increased after being treated for 4 weeks ( P 〉 0. 05). Conclusions The decreased myocardial calsarcin-1 level participates in cardiac hypertrophy development, Telmisartan increases myocardial Calsarcin-1 level expression, which may be one of its mechanisms in reversing cardiac hypertrophy.
