流式细胞术检测慢性再生障碍性贫血患者T细胞活化的研究

摘　　要：[目的]探讨T细胞活化在再生障碍性贫血(aplastic anemia,AA)发病机制中的作用。[方法]通过流式细胞术检测CD25/CD3^+CD4^+、CD69^+/CD3^+CD4^+、HLA-DR/CD3^+CD4^+、CD25^+/CD3^+CD8^+、CD69^+/CD3^+CD8^+、HLA-DR/CD3^+CD8^+T细胞在慢性AA组患者外周血中的表达水平,并取61例患者为AA组与30例非血液系统疾病对照人群为对照组予比较。[结果]AA组CD25^+/CD3^+CD4^+T细胞比例为8.04%,对照组为6.75%,P<0.05,有统计学意义。AA组HLA-DR/CD3^+CD8^+T细胞比例为15.90%,显著高于对照组6.40%(P<0.01)。[结论]T细胞异常活化可能在再障发病或病情进展过程中发挥重要作用。[Objective] To investigate the expression of CD25＋/CD3＋CD4% CD69＋/CD3＋CD4% HLA-DR/CD3＋CD4＋ CD25＋/CD3＋CD8, ＋CD＋9＋/CD3＋CD8% HLA-DR/CD3＋CD8＋ T lymphocytes activation markers in peripheral blood of patients of aplastic anemia（AA） and to discuss their roles of pathogenesis of AA.[Methods] The ratio of T cells activation in peripheral blood in ftl patients with AA was detected by flow cytometry（FCM）, and compared with the normal control group. [Results] The percentage of peripheral blood CD25＋/CD3＋CD4＋ T cells（8.04%）in AA group was significantly higher than that（6.75%）in the normal control group（P〈0.05）. There was a significant difference according to the ratio of HLA-DR/CD3＋CD8＋ T cells in peripheral blood between AA group （15.90%） and the control group（ft. 40%）. And there was no significant difference of CD69＋/CD3＋CD4% HLA-DR/CD3＋CD4% CD25＋/CD3＋ CD8＋, CDft9＋/CD3＋CD8＋T cells between AA group and the control group. [Conclusion] The increased activation of CD25＋/CD3＇CD4＋ and HLA-DR/CD3＋CD8＋ T cells might play a major role in the pathogenesis of AA.
