摘  要:目的:探讨清心开窍方多糖成分对Aβ25-35杏仁核注射诱导AD大鼠脑组织凋亡信号传导分子Caspase-3及凋亡相关基因Bax、Bcl-2表达的影响。方法:雄性SD大鼠40只,随机分为正常组、模型组、安理申组、清心开窍方组和多糖组,每组8只。采用双侧杏仁核注射Aβ25-35制造AD大鼠模型,造模后第2天起及灌胃给药2周后,观察大鼠水迷宫空间记忆能力,免疫组化方法研究脑组织Caspase-3、Bax、Bcl-2及βAPP的表达。结果:中药各组能明显改善AD大鼠学习记忆能力(P<0.05,P<0.01);清心开窍方组、多糖组大鼠大脑皮层、海马区Bax、Caspase-3、βAPP表达水平降低(P<0.05,P<0.01)。结论:清心开窍方能显著改善AD大鼠学习记忆的能力,可能是通过降低皮质、海马Bax、Caspase-3、βAPP表达而发挥作用。Objective:To study effect of qing xin kai-qiao-fang and polysaccharide on the expression of the Apoptosis Signal Transtuduetion Molecule Caspase-3 and Apoptosis Associated Gens Bax、Bcl-2 in rat models of dementia induced by bilateral injection of beta-amyloid 25-35 in amygdala.Methods:Forty male SD rats were randomly divided into five groups: the control group,the model group,the Aricept group,the qing xin kai-qiao-fang group and the polysaccharide group,with 8 rats in each group.AD was induced by Aβ25-35 injected into the bilateral amygdala to observe the spatial learning and memony ability,the expression of Bax、Bcl-2、Caspase-3 and βAPP by immunohistochemistry in the brain of the animal,Molding the very next day start respectively,medicine filling after 2 weeks.Results:Learning and memory ability of qing xin kai-qiao-fang groups is improved,the expression of Bax、Caspase-3、andβAPP decreased in the cortex and hippocampus(P0.05~0.01).Conclusion:Qing xin kai-qiao-fang may obviously improved in learning and memory ability of rats with AD by decreasing the expression of Bax、Caspase-3 and βAPP in the cortex and hippocampus.
