摘  要:目的建立一种检测慢性粒细胞白血病的方法。方法此实验在已经筛选出慢性粒细胞白血病(CML)K562细胞5个特异性、结合率较高的核酸适体的基础上,构建了一种检测方法:采用两种纳米金属材料分别标记5个适体,将标记后的10个适体,两两组合与CML K562同时结合,一个作为捕获分子,一个作为测定分子与CML K562结合后,经氧化还原反应将测定分子上的金属转化成离子状态,利用电化学工作站检测对应的电流。结果筛选出了最佳组合(P2-S3-P1-S5)实现对CML K562的检测,得到了检测细胞数目与DPV电流值的拟合曲线方程。结论得到一种检测下限可达到50个细胞的诊断慢性粒细胞白血病的电化学生物传感器。Objective To creat a method of detection for chronic myelocytic leukemia. Methods A method of detection had been built, based on an experiment in lab, during which, 5 highly-combined aptamers of CML K562 cells had been se- lected. Five aptamers were marked with two kinds of nanoparticles separately. The ten marked aptamers were divided into 20 pairs by every two combination. In every pair, the aptamers, one as the capture molecules, the other the determination molecules, were combined with CML K562 cells. Then the Au atoms were transformed into the state of An-ion by REDOX reactions and electric current was sensed by electrochemical detection. Results The best pair of aptamers was selected to detecte CML K562, the fitted curve equation to test cells number and DPV of current value was obtained. Conclusion Get a electrochemical biosensor which can be used to diagnose CML K562 cells, with a detection limit up to 50 cells.
