DNA裂解因子-45在宫颈癌中的表达及与HPV感染的关系

摘　　要：目的探讨DFF45蛋白在宫颈上皮内瘤变(CIN)及宫颈癌(CC)组织中的表达及其临床意义,分析DFF45蛋白的表达和HPV16/18感染的关系。方法用免疫组化方法检测DFF45蛋白在正常宫颈组织、CIN组织及CC组织中的表达情况。结果 DFF45在正常宫颈组织、CIN组织和宫颈癌组织中的表达呈递增趋势(χ2=18.65,P<0.01)。DFF45蛋白在CIN I中的表达低于CINⅡ-Ⅲ(P<0.01);DFF45的表达改变与临床分期及HPV感染有关(P<0.05),与年龄、组织类型、分化程度及淋巴结转移无关。结论 DFF45蛋白的表达与HPV的感染密切相关,DFF45和HPV相互协调、相互促进,促进了宫颈癌的发生和进展。DFF45蛋白具有抑制凋亡的作用,有望成为一抗癌基因治疗的作用靶点。Objective To explore the expression and its significance of DNA fragmentation factor-45 in normal cervical tissues,cervical intraepithelial neoplasms and cervical cancer. To analysis the relationship between the expression of DFF45 and the infection of HPV16/18. Methods The expressions of DFF45 in normal cervical tissues （15cases） and cervical intraepithelial neoplasms （48 cases） and cervical cancer tis sues （41 cases） were detected by immunohistochemical technique. Results The expressions of DFF45 were significantly different between the tissues of normal cervical tissues and cervical intraepithelial neoplasms and cervical cancer （ P〈0.05）. The expressions of DFF45 were significantly different between the tis sues of CINI and CINII-Ⅲ. No statistical difference was found in expression of DFF45 between normal cer vical tissues and CINI. The expressions of DFF45 were significantly different between clinical stage I and stage II, significantly different was found in patient with and without HPV infection. No statistical differ ence was found in expression of DFF45 in different histological type, histological degree, lymph node me tastasis and age. Conclusion The expression of DFF45 protein is closely related with the infection of HPV,DFF45 and HPV coordination and promote each other, promote cervical cancer occurrence and progress. DFF45 protein can inhibit apoptosis ,which is expected to be a gene therapy target of cancer.
