凋膜止崩液宫腔靶向给药对ADUB模型大鼠子宫内膜组织中MMP-1、TIMP-1、ER及亚型表达的影响

摘　　要：目的探讨凋膜止崩液宫腔靶向给药治疗无排卵功血(ADUB)子宫内膜增生症的可能机制,为临床治疗ADUB提供一种新方法。方法卵巢去势后利用雌激素负荷法建立ADUB子宫内膜增生症大鼠模型,凋膜止崩液宫腔内给药后处死,留取子宫标本,采用RT-PCR检测法测定各组大鼠子宫内膜组织中基质金属蛋白酶-1(MMP-1)和基质金属蛋白酶抑制因子-1(TIMP-1)基因表达水平的变化;采用免疫组化技术检测各组大鼠子宫内膜组织中雌激素受体(estrogenreceptor,ER)、ERα、β亚型的表达变化。结果①模型对照组与空白组比较,MMP-1mRNA表达明显升高,TIMP-1mRNA表达降低,差异有统计学意义(P<0.05)。实验大、小剂量组与模型对照组比较,MMP-1mRNA表达降低,TIMP-1mRNA表达升高,差异有统计学意义(P<0.05),而大小剂量组间比较无显著性差异(P>0.05)。②模型对照组与空白组比较,ER、ERα、β表达明显升高,差异有统计学意义(P<0.05)。实验大、小剂量组与模型对照组比较,ER、ERα、β表达下调,差异有统计学意义(P<0.05),而实验大、小剂量组间比较无显著性差异(P>0.05)。结论凋膜止崩液宫腔靶向给药后ADUB模型大鼠子宫内膜组织中MMP-1mRNA表达下降,TIMP-1mRNA表达升高,ER、ERα、β表达下调,可能是其治疗子宫内膜增生症的机制。Objective To explore the mechanism of Diaomozhibeng liquid for treatment of endometrium hyperplasia （EH） and to provide a new method of clinically treating endometrial hyperplasia. Methods Rat model was induced by ovariectomy combining with estrogen. The expression levels MMP - 1 and TIMP - 1 were checked by real - time PCR and ER, ERa, ER~ were checked by immunohistochemistry. Results Compared with the control group, the expression of MMP - lmRNA in model group was in- creased,while that of TIMP- 1mRNA was decreased. The difference was statistically significant （ P 〈 0.05 ）. Compared with model group, the large dose of Diaomozhibeng liquid and small dose of Diaomozhibeng liquid could decrease the expression of MMP - lmRNA and increase the expression of TIMP - 1mRNA, and the difference was statistically significant （ P 〈 0.05 ）. While compared the large dose of Diaomozhibeng liquid with small dose of Diaomozhibeng liquid, there had no significant difference （ P 〉 0.05 ）. Compared with the control group, the expression of ER, ERa, β in model group was significant increased （ P 〈 0.05 ）. Compared with model group, the big dose of Diaomozhibeng liquid and small dose of Diaomozhibeng liquid, the expression of ER, ERa, β decreased. The difference was statistically significant （ P 〈 0.05 ）. There had no significantly different between the large dose of Diaomozhibeng liquid and small dose of Diaomozhibeng liquid（ P 〉 0.05 ）. Conclusion The mechanism of treatment for endometrial hyperplasia with Diaomozhibeng liquid uterine cavity target intervention may be associated with reducing the expression of MMP - 1mRNA ;ER, ERa, β and increasing the expression of TIMP - 1 mRNA .
